为探讨ADAMTS1基因遗传多态性对大白猪、杜洛克猪、豫南黑猪3个品种繁殖性状的影响,采用PCR-RFLP方法在大白猪、杜洛克猪、豫南黑猪中检测猪ADAMTS1基因的多态性分布,并进行基因型与猪繁殖性能的关联性分析。结果显示,ADAMTS1外显子7上G72A由碱基G突变为A,属同义突变,在3个品种群体中多态位点均以等位基因A基因频率最高;AB基因型占总基因型的46%,高于AA基因型(40%)与BB基因型(14%),呈现AB>AA>BB的趋势;该位点BB型个体的总产仔数、产活仔数、初生窝重、21日龄窝重高于AA型和AB型。结果表明,ADAMTS1基因多态位点与母猪繁殖性能显著相关,可以用于猪分子遗传育种的候选基因。

In order to find the molecular marker used in marker-assisted selection(MAS) to accelerate pig breed-ing progress,the pig A disintesrin and metallo-proteinase with thrombospondin motifs-1(ADAMTS1) was selected to study the association between its polymorphism and reproductive traits of Yorkshire,Duroc,Yunan black pig by PCR-RFLP and DNA sequencing methods.The results showed that the PCR products had polymorphism.Three genotypes(AA,AB,and BB),and one single nucleotide mutation(G→A) was detected in three pig breeds,and this mutation resulted in no amino acid change.The highest gene frequency was allele A. The genotype frequencies of AA,AB,BB of the G72A site were 40%,46% and 14% respectively.In the site of G72A,AA genotype had higher TNB,NBA,LBW,number of weaning and litter weight at 21 days than that of AB,BB.Therefore,these results suggest that the ADAMTS1 is a candidate gene that affects reproductive traits in pigs.
