基于冬虫夏草全基因组测序,对其HSP70基因测序技术进行分析,以5种虫草样本为试材,比较冬虫夏草HSP70基因整序列PCR测序技术与近缘种HSP70基因分段PCR测序技术两种方法适用性。结果表明,前者进程较短,效率较高,但仅适用于冬虫夏草材料;而后者通用性较好,适用于虫草多物种的分析,但工作量较大。HSP70基因在冬虫夏草种内分化小于种间的分化,冬虫夏草种及其近缘种HSP70基因序列分析结果与Sung的虫草分类系统分类相吻合。
基于鼕蟲夏草全基因組測序,對其HSP70基因測序技術進行分析,以5種蟲草樣本為試材,比較鼕蟲夏草HSP70基因整序列PCR測序技術與近緣種HSP70基因分段PCR測序技術兩種方法適用性。結果錶明,前者進程較短,效率較高,但僅適用于鼕蟲夏草材料;而後者通用性較好,適用于蟲草多物種的分析,但工作量較大。HSP70基因在鼕蟲夏草種內分化小于種間的分化,鼕蟲夏草種及其近緣種HSP70基因序列分析結果與Sung的蟲草分類繫統分類相吻閤。
기우동충하초전기인조측서,대기HSP70기인측서기술진행분석,이5충충초양본위시재,비교동충하초HSP70기인정서렬PCR측서기술여근연충HSP70기인분단PCR측서기술량충방법괄용성。결과표명,전자진정교단,효솔교고,단부괄용우동충하초재료;이후자통용성교호,괄용우충초다물충적분석,단공작량교대。HSP70기인재동충하초충내분화소우충간적분화,동충하초충급기근연충HSP70기인서렬분석결과여Sung적충초분류계통분류상문합。
Based on the entire genome sequencing of Ophiocordyceps sinensis by using samples of 5 Cordyceps spe-cies, two methods of sequencing HSP 70 gene were analyzed .One was directly PCR sequencing of HSP 70 gene of O.sinensis, and the other was PCR three fragments of HSP 70 gene of its allied species.The results showed that the former had a shorter process with higher efficient , and only limited to O.sinensis, while the later could be ap-plied to many species of Cordyceps, but the workload was relatively heavy .The intraspecific differentiation of HSP70 gene in O.sinensis was less than that of its allied species .The phylogenetic analysis of HSP 70 gene on O.sinensis and its allied species was consistent with Sung ’ s system of Cordyceps.
