目的:探讨黄芪甲苷(ASIV)对脂多糖(LPS)诱导急性肺损伤(ALI)的保护作用,并初步探讨其抑制内质网应激介导的细胞凋亡的作用。方法:随机将50只SD大鼠分成5组,每组10只。对照组(静注生理盐水1mg/kg),ALI组(静注LPS,5mg/kg), ASIV组(注射LPS前,给药3、6、12mg/kg ASIV连续灌胃7天)。观察注射12h后肺组织湿干重(W/D)、western blotting检测葡萄糖调节蛋白78(GRP78)、C/EBP环磷酸腺苷反应元件结合转录因子同源蛋白(CHOP)的蛋白表达。结果:ALI组W/D值较对照组上升(P<0.01),GRP78、ChOP蛋白表达较对照组升高(P<0.01);ASIV组W/D值较ALI组下降(P<0.01),GRP78、ChOP蛋白表达较ALI组降低(P<0.01),并呈一定的剂量依赖性。结论:黄芪甲苷对急性肺损伤大鼠有保护作用,其作用机制可能与内质网应激有关。

목적:탐토황기갑감(ASIV)대지다당(LPS)유도급성폐손상(ALI)적보호작용,병초보탐토기억제내질망응격개도적세포조망적작용。방법:수궤장50지SD대서분성5조,매조10지。대조조(정주생리염수1mg/kg),ALI조(정주LPS,5mg/kg), ASIV조(주사LPS전,급약3、6、12mg/kg ASIV련속관위7천)。관찰주사12h후폐조직습간중(W/D)、western blotting검측포도당조절단백78(GRP78)、C/EBP배린산선감반응원건결합전록인자동원단백(CHOP)적단백표체。결과:ALI조W/D치교대조조상승(P<0.01),GRP78、ChOP단백표체교대조조승고(P<0.01);ASIV조W/D치교ALI조하강(P<0.01),GRP78、ChOP단백표체교ALI조강저(P<0.01),병정일정적제량의뢰성。결론:황기갑감대급성폐손상대서유보호작용,기작용궤제가능여내질망응격유관。
Objective To explore the effect of Astragaloside IV on apoptosis mediated by endouplasmic reticulum stress (ERS)in a rat model of acute lung injury (ALI)induced by lipopolysaccharide (LPS). Methods Fifty SD rats were randomly divided into con-trol group,ALI group,and ASIV group,10 in each group. The rats in the control group were treated with normal saline. The rats in the ALI group were treated with LPS (5mg/kg). The rats in the ASIV groups were pretreated with ASIV (3,6,12mg/kg)for 7days.After 12h injection,the change of wet -dry weight (W/D)were determined. The expressions of glucose -regulated protein 78 (GRP78)and C/EBP homologous protein (CHOP)were determined by Western blot. Results Compar2ed with the control group,the W/D was increased (P#0.01),the expression ofGRP78 and CHOP were increased in the ALI group (P#0.01). Compared with ALI group,the W/D was decreased (P # 0.01 ),the expression of GRP78 and CHOP were decreased in the ASIV groups (P # 0.01). Conclusion Astragaloside IV has a protective effect on LPS-induced acute lung injury,which is partially via endouplasmic reticulum stress.
