目的:研究舒脉汤对血管内皮细胞生成因子(VEGF)、碱性成纤维细胞生成因子(bFGF)的影响及其可能参与的信号通路。方法原代培养人脐静脉内皮细胞,以舒脉汤刺激内皮细胞,部分给予 PI3K 抑制剂,LY294002预处理,收集细胞测定 VEGF、bFGF 基因表达,收集上清液测定 VEGF、bFGF 蛋白表达。结果舒脉汤可以明显增加人脐静脉内皮细胞 VEGF、bFGF 基因及蛋白表达,且这一过程可被 LY294002所阻断。结论舒脉汤通过上调内皮细胞 VEGF、bFGF 表达来发挥促血管新生作用,PI3K 信号通路参与此过程。

목적:연구서맥탕대혈관내피세포생성인자(VEGF)、감성성섬유세포생성인자(bFGF)적영향급기가능삼여적신호통로。방법원대배양인제정맥내피세포,이서맥탕자격내피세포,부분급여 PI3K 억제제,LY294002예처리,수집세포측정 VEGF、bFGF 기인표체,수집상청액측정 VEGF、bFGF 단백표체。결과서맥탕가이명현증가인제정맥내피세포 VEGF、bFGF 기인급단백표체,차저일과정가피 LY294002소조단。결론서맥탕통과상조내피세포 VEGF、bFGF 표체래발휘촉혈관신생작용,PI3K 신호통로삼여차과정。
Objective To study the effect of Shumaitang on vascular endothelial growth factor(VEGF)and basic fi-broblast growth factor(bFGF)expression of vascular endothelial cells and its possible signal pathway. Methods Primary cultured human umbilical vein endothelial cells(HUVEC)were stimulated by Shumaitang,part of them were pretreated by PI3K inhibitor LY294002. Cells were collected for determination of VEGF,bFGF gene expression;the supernatant was collected for determination of VEGF,bFGF protein expression. Results Shumaitang could increase the gene and protein expression of VEGF and bFGF of vascular endothelial cells significantly,and this effect was blocked by LY294002. Conclusion PI3K/VEGF and PI3K/ bFGF may take part in the Shumaitang-mediated-angiogenesis.
