目的:建立能够实现甲基化敏感扩增多态性(MSAP)荧光检测初始数据自动分析的方法。方法:以17周龄的北京油鸡为实验材料,应用荧光标记的MSAP方法对肌肉组织基因组DNA甲基化水平进行检测,对获得的MSAP荧光检测初始数据通过直接取整法、整区间放置法和半区间放置法等3种计算机处理方法进行分析和比较。结果:对于MSAP荧光检测初始数据的自动转换,半区间放置法与标准的人工转换方法最接近(P>0.05),整区间放置法次之(P>0.05),而直接取整法与标准的人工转换方法差异最大,达到显著水平(P<0.05)。结论:整区间放置法和半区间放置法适用于MSAP荧光检测初始数据的自动分析,建立了基于半区间放置法的在线自动分析软件MSAP Analyst,可实现MSAP荧光检测初始数据的自动分析。

Objective: To built a suitable automatic analysis system based on compute to process the initial da-ta of methylation level detected by fluorescence labeled methylation sensitive amplified polymorphism(MSAP). Methods: With fluorescently labeled MSAP, the DNA methylation levels in muscle tissues from 17-week-old Bei-jing You chicken were detected. The initial data of MSAP were analyzed using manual processing usually as a standard solution, and three automatic processing systems based on compute: direct integral method, whole interval method and half interval method. Results: In the analysis of initial data of methylation level detected by fluores-cence labeled MSAP, no differences was found between manual processing and whole interval method(P>0.05) or half interval method(P>0.05), and the half interval method is more near to manual processing than whole interval method, but significant difference appeared between manual processing and direct integral method(P<0.05). Conclu-sion: The whole interval and half interval methods as a substitute for manual processing can be used to realize the automatic analysis of initial data of methylation level detected by fluorescence labeled MSAP, and one new on-line software named MSAP Analyst based on half interval method was built.
