大量高质量且能再生的原生质体是真菌遗传转化、基因修饰的重要保证。本试验选用苹果腐烂病菌强致病力菌株SDAU11-175,从酶种类、菌丝年龄、酶解时间、渗透压稳定剂及再生培养基五个方面对原生质体制备及再生条件进行了优化。结果显示:获得苹果腐烂病菌原生质体最大产量的条件是菌丝年龄54 h、3%崩溃酶和3%裂解酶组合、0.7 mol/L NaCl作为渗透压稳定剂、28℃酶解3 h,所得原生质体在YPS培养基上再生效果最好,再生率可达42.21%。
大量高質量且能再生的原生質體是真菌遺傳轉化、基因脩飾的重要保證。本試驗選用蘋果腐爛病菌彊緻病力菌株SDAU11-175,從酶種類、菌絲年齡、酶解時間、滲透壓穩定劑及再生培養基五箇方麵對原生質體製備及再生條件進行瞭優化。結果顯示:穫得蘋果腐爛病菌原生質體最大產量的條件是菌絲年齡54 h、3%崩潰酶和3%裂解酶組閤、0.7 mol/L NaCl作為滲透壓穩定劑、28℃酶解3 h,所得原生質體在YPS培養基上再生效果最好,再生率可達42.21%。
대량고질량차능재생적원생질체시진균유전전화、기인수식적중요보증。본시험선용평과부란병균강치병력균주SDAU11-175,종매충류、균사년령、매해시간、삼투압은정제급재생배양기오개방면대원생질체제비급재생조건진행료우화。결과현시:획득평과부란병균원생질체최대산량적조건시균사년령54 h、3%붕궤매화3%렬해매조합、0.7 mol/L NaCl작위삼투압은정제、28℃매해3 h,소득원생질체재YPS배양기상재생효과최호,재생솔가체42.21%。
Protoplasts with high quality and high regeneration rate are essential in genetic transformation and gene modification of fungi .Using highly pathogenic Valsa mali var.mali strain SDAU11-175 as research object , the protoplast preparation and regeneration conditions were optimized from five aspects of enzyme types, mycelia age, enzymolysis time, osmotic stabilizer and regeneration medium .The results showed that the conditions to obtain the maximum protoplast output were culturing mycelia for 54 hours and treating them with 3%driselase and 3% lysing enzymes for 3 hours at 28℃ using 0 .7 mol/L NaCl as osmotic stabilizer . The protoplasts regenerated best on YPS medium with the regeneration rate as 42 .21%.
