目的:初步研究罗汉松种子提取物对人胃癌细胞MGC-803的抑制作用。方法采用噻唑蓝(MTT)法测定不同浓度梯度的罗汉松种子提取物作用人胃癌MGC-803细胞24,48,72 h的增殖抑制率。采用RT-PCR检测MGC-803细胞凋亡相关基因bcl-2和p53 mRNA的表达。结果不同质量浓度罗汉松种子提取物对MGC-803细胞均有增殖抑制作用,且随着质量浓度的增加和作用时间的延长,抑制率呈上升趋势。RT-PCR结果显示,不同质量浓度罗汉松种子提取物作用于MGC-803细胞,bcl-2 mRNA水平随质量浓度的增加而明显下降,而p53 mRNA水平则随质量浓度的增加而显著增强。结论罗汉松种子提取物对人胃癌细胞MGC-803有明显的体外抑制作用,其抑制作用与药物的质量浓度和作用时间呈正相关,可能与其调控相关凋亡基因的表达有关。

목적:초보연구라한송충자제취물대인위암세포MGC-803적억제작용。방법채용새서람(MTT)법측정불동농도제도적라한송충자제취물작용인위암MGC-803세포24,48,72 h적증식억제솔。채용RT-PCR검측MGC-803세포조망상관기인bcl-2화p53 mRNA적표체。결과불동질량농도라한송충자제취물대MGC-803세포균유증식억제작용,차수착질량농도적증가화작용시간적연장,억제솔정상승추세。RT-PCR결과현시,불동질량농도라한송충자제취물작용우MGC-803세포,bcl-2 mRNA수평수질량농도적증가이명현하강,이p53 mRNA수평칙수질량농도적증가이현저증강。결론라한송충자제취물대인위암세포MGC-803유명현적체외억제작용,기억제작용여약물적질량농도화작용시간정정상관,가능여기조공상관조망기인적표체유관。
Objective To preliminarily study the inhibition effect of the podocarpus seed extract on human gastric cancer cell MGC-803. Methods The methyl thiazolyl tetrazolium ( MTT ) assay was adopted to detect the the 24, 48, 72 h proliferation inhibition rate of human gastric cancer MGC-803 cells caused by different concentrations gradient of the podocarpus seed extract. RT-PCR was adopted to detect the expression of MGC-803 apoptosis-related genes P53 and BcL-2 mRNA. Results Different concentrations of podocarpus seed extract had the proliferation inhibiting effect on MGC-803 cells, moreover as the increase of podocarpus seed extract concentration and the extension of the treatment time, the inhibition rate showed the upward trend. The RT-PCR results showed that in different concentrations of podocarpus seed extract acting on the MGC-803 cells, the expression level of BcL-2 mRNA was decreased with the concentration increase, while the expression level of P53 mRNA was significantly enhanced with the concentration increase. Conclusion Podocarpus seed extract has significantly inhibiting effect in vitro on human gastric cancer cell MGC-803, and its inhibition effect is positively correlated with the drug concentration and the action time, which may be related to the regulation of the apoptosis-related genes expression.
