目的:研究 EphA2基因在大鼠白内障晶状体中的表达水平变化的情况。方法选用18只SD大鼠的36个晶状体,分为3组,每组6只,分别为2月龄透明晶状体组(青年组)、12月龄透明晶状体组(老年组)和12月龄白内障晶状体组(白内障组)。采用实时荧光定量 PCR 和Western blot检测青年组、老年组和白内障组晶状体中EphA2基因和蛋白的表达水平。结果 Western blot检测结果显示EphA2蛋白在白内障组中表达水平明显低于老年组(t=10.952,P=0.000);而老年组比青年组EphA2蛋白表达水平呈轻度的升高(t=-4.456,P=0.001)。Q-PCR结果显示,白内障组中的EphA2 mRNA表达水平较老年组呈明显的升高(t=-4.518,P=0.001);老年组中的EphA2 mRNA表达水平较青年组呈轻度的降低(t=8.237,P=0.000)。结论 EphA2蛋白表达水平下降与大鼠白内障发生密切相关。随着大鼠年龄增加,EphA2蛋白表达水平呈轻度的升高。

Objective To investigate the relationship between expression of EphA2 and cataract in rat. Methods Thirty-six lenses from SD rats were allocated into three groups:clear younger, older and cataractous lenses. Expression of EphA2 in the lenses of younger, older, and cataractous was detected by quantitative real time RT-PCR and western blot. Results Western blot analysis showed that EphA2 protein in rat cataractous lenses was lower compared to clear older lenses (t=10.952, P=0.000). The older lenses slightly higher in the level expression of EphA2 protein compared to younger lenses (t=-4.456, P=0.001). Real time RT-PCR analysis showed that EphA2 mRNA expression in rat cataractous lenses was higher than clear older lenses (t=-4.518, P=0.001). However, EphA2 mRNA expression in older lenses was lower than younger lenses (t=8.237, P=0.000). Conclusions Lower level expression of EphA2 was related to cataract in rats. The expression level of EphA2 protein in rats’ lenses was affected by age.
