目的:探讨异氟烷(Isoflurane)对新生大鼠脑室管膜下区神经干细胞的影响。方法将出生后5 d 的20只新生大鼠随机分为对照组(Control 组)、0.5%异氟烷组(ISO 0.5%组)、0.75%异氟烷组(ISO 0.75%)和1.5%异氟烷组(ISO 1.5%)组。免疫荧光方法标记脑室管膜下区的神经干细胞,观察异氟烷作用后神经干细胞凋亡以及增殖情况的变化。结果0.5%、0.75%和1.5%浓度异氟烷可导致新生大鼠室管膜下区神经干细胞增殖减少(P <0.05),而对神经干细胞的凋亡没有影响。结论异氟烷浓度依赖性导致新生大鼠室管膜下区神经干细胞增殖减少,而并不影响神经干细胞的凋亡。

목적:탐토이불완(Isoflurane)대신생대서뇌실관막하구신경간세포적영향。방법장출생후5 d 적20지신생대서수궤분위대조조(Control 조)、0.5%이불완조(ISO 0.5%조)、0.75%이불완조(ISO 0.75%)화1.5%이불완조(ISO 1.5%)조。면역형광방법표기뇌실관막하구적신경간세포,관찰이불완작용후신경간세포조망이급증식정황적변화。결과0.5%、0.75%화1.5%농도이불완가도치신생대서실관막하구신경간세포증식감소(P <0.05),이대신경간세포적조망몰유영향。결론이불완농도의뢰성도치신생대서실관막하구신경간세포증식감소,이병불영향신경간세포적조망。
Objective To investigate the effects of isoflurane(ISO)on the neural stem cell in the subventricular zone of neonatal rats.Methods Twenty rats on postnatal day 5 were randomly divided in-to 4 groups and exposed to varying ISO concentrations(0%,0.5%,0.75%and 1.5%)respectively.Immu-nofluorescence technique was used to label and detect apoptosis and proliferation of the neural stem cells in the subventricular zone.Results The proliferation of neural stem cells in the subventricular zone was sig-nificantly decreased in rats with 0.5%,0.75% and 1.5% ISO exposure(P <0.05)and no effects on ap-optosis were observed between the groups.Conclusion In neonatal rats,isoflurane decreases neural stem cell proliferation of the subventricular zone in a concentration dependent manner and it does not affect ap-optosis.
