采用均匀设计法优化了达托霉素发酵培养基配方,并对30 L 罐生产工艺进行了优化。确定达托霉素最优发酵培养基配方为:葡萄糖2.0%、淀粉2.0%、冷榨豆饼粉0.5%、蛋白胨0.6%;以该配方进行30 L 发酵罐生产,当前体癸酸钠的补料起始时间为发酵开始28 h、补料总量为1.5%时,最终发酵效价提高了81%。对达托霉素的工业化生产具有重要的指导意义。
採用均勻設計法優化瞭達託黴素髮酵培養基配方,併對30 L 罐生產工藝進行瞭優化。確定達託黴素最優髮酵培養基配方為:葡萄糖2.0%、澱粉2.0%、冷榨豆餅粉0.5%、蛋白胨0.6%;以該配方進行30 L 髮酵罐生產,噹前體癸痠鈉的補料起始時間為髮酵開始28 h、補料總量為1.5%時,最終髮酵效價提高瞭81%。對達託黴素的工業化生產具有重要的指導意義。

Uniform design was used to optimize the medium of daptomycin shaking flask fermentation.The process was also optimized in 30 L fermentor.The optimal fermentation medium was obtained as follows:glu-cose 2.0%,starch 2.0%,soybean flour 0.5%,peptone 0.6%.The fermentation process of daptomycin in 30 L fermentor with above optimum fermentation medium was optimized as follows:the initial feeding time of precur-son sodium caprate was at 28 h,the total feeding amount was 1.5%.The final fermentation potency was im-proved by 81%,which was significant in industrial production of daptomycin.
