为探究冠突散囊菌与veA基因相关的差异表达蛋白,利用双向电泳-质谱技术对冠突散囊菌野生型菌株只产生有性孢子时期、△veA缺失菌株只产生无性孢子时期、△veA缺失菌株既产生无性孢子又产生有性孢子时期的蛋白进行分离和鉴定。结果表明:经检测得到107个功能各异的蛋白点,其中,大多数蛋白与代谢相关,最多的是具有氧化还原功能的蛋白。3种蛋白与产孢和发育有关,包括α-微管蛋白、蛋白磷酸酶2A、真核细胞翻译起始因子3。说明,用双向电泳-质谱技术可以有效地分析冠突散囊菌与veA基因相关的差异蛋白,有助于认识冠突散囊菌的产孢机理。

To explore the relationship of veA gene and the differentially expressed proteins of E.cristatum,three protein samples of E.cristatum of only producing sexual spore period in wild type strain,only producing asexual spore period in△veA deletion type strain and producing both asexual spores and sexual spores in △veA deletion type strain were collected.Two-dimensional electrophoresis was used to isolate the proteins,and the differential protein spots are identified by mass spectrometry.Results:One hundred and seven differential proteins were identified by mass spectrometry including three proteins may related to spore and development:tubulin alpha,protein phosphotase 2A,eukaryotic translation initiation factor 3.The majority of these proteins were related to metabolism and most of them were oxidordeuctase. Results showed that two-dimensional electrophoresis combined with mass spectrometry could be used to analyze the differentially expressed proteins related to veA gene of E.cristatum,helping to understand spore production mechanism of E.cristatum.
