目的:检测复发性口腔溃疡(ROU)患者体内外周血IL-12的表达及辅助性T细胞(Th)表面白细胞介素12受体(IL-12R)的表达情况,探讨IL-12及其受体对ROU患者的Th细胞的免疫失衡产生的影响。方法分离滨州医学院附属医院20例ROU患者和健康人的外周血单个核细胞,采用流式细胞术测定CD4+T细胞上IL-12R的表达量,ELISA检测外周血IL-12的表达水平。结果 ROU患者Th细胞表面的IL-12R表达水平高于正常对照组,ROU患者外周血IL-12的表达低于正常对照组,两结果均具有统计学意义(P<0.05)。结论 ROU 患者体内免疫紊乱, IL-12及其受体的异常表达与疾病相关,为ROU患者的治疗寻找新的干预手段提供了理论依据。

Objective To detect the concentration of the plasma interleukin 12 (IL-12) and the expression of interleukin 12 receptor (IL-12R) on helper T cell in recurrent oral ulcer (ROU), and investigate the influence of IL-12 and IL-12R to Th cell immune imbalance in patients with ROU. MethodsThe peripheral blood was obtained from 20 patients in active stage of ROU and 20 healthy controls. Flow cytometry was used to detect the quantity of IL-12R in CD4+T cells in the blood, ELISA was used to detect the level of IL-12.Results The expression of IL-12R on CD4+T cells was higher in the patients with ROU (P<0.05) and IL-12 content was significantly lower than those in the healthy controls (P<0.05).Conclusion This study demonstrates that IL-12R expression level on CD4+ T cells and IL-12 in peripheral blood of ROU patients are both significantly increased, that provide novel evidence for the therapeutic approach of ROU.
