目的:观察白藜芦醇( RES)干预对快速起搏右心房诱发的持续性心房颤动( AF)猪超氧化物歧化酶( SOD)活性和丙二醛( MDA)含量的影响,探讨RES对持续性AF时氧化损伤的抑制作用。方法18只小家猪(雌雄不拘)按完全随机设计的随机分组方法(采用动物编号和随机分组表)分为起搏组(ATP组,n=6)、假手术组(Sham组,n=6)和RES干预组(n=6),采用Seldinger血管穿刺技术穿刺右或左侧颈内静脉送入双极电极至右心房并连接至实验用起搏器,快速起搏心房(500次/min)2周(Sham组猪不起搏)制备持续性AF实验模型。 RES组猪于起搏前一周开始服用RES (2.5 mg?kg-1?d-1)。用ELISA法检测3组手术前后MDA、SOD水平。结果(1)起搏前:MDA、SOD值在3组间差异均无统计学意义(均为P﹥0.05)。(2)起搏2周后:①与起搏前比较,ATP组和RES组MDA明显升高,SOD降低(均为P﹤0.01);Sham组的差异无统计学意义(均为P﹥0.05);②组间比较, ATP组MDA值明显高于Sham组和RES组(均为P﹤0.01),RES组MDA值稍高于Sham组,但差异无统计学意义(P﹥0.05);ATP组SOD值明显低于Sham组和RES组(均为P﹤0.01),RES组SOD值明显高于ATP组(P﹤0.01),与Sham组比较差异无统计学意义(P﹥0.05)。结论快速起搏右心房诱导的持续性AF猪的氧化应激反应程度明显增高,另外RES可明显减轻持续性AF猪的氧化应激反应。

Objective To observe the effect of resveratrol on superoxide dismutase( SOD) activity and maleic dialdehyde ( MDA) content in pigs with sustained atrial fibrillation induced by right tachypacing and investigate the oxidative damage inhibition of resveratrol in sustained atrial fibrillation. Methods Eighteen pigs were randomly divided into three groups for atrial tachypacing ( ATP ) group ( n = 6 ) , resveratrol-intervened (RES) group (n=6) and sham group (n=6) in completely randomized design. The pigs in three groups were all put the pacemakers ( AOO) , only the pigs in ATP group and RES group were paced at 500 beats per minute for 2 weeks. Resveratrol (2. 5 mg?kg-1?d-1 ) were given orally in 3 divided doses, started 7 days prior to ATP onset and continued until the morning of the final open-chest experiment. SOD and MDA content were tested by ELISA. Results (1) Before atrial tachypacing (ATP), MDA and SOD in all groups were similar (all P﹥0. 05). (2) After 2 weeks of ATP:① compared with those before ATP in each group, ATP group and RES group changed( MDA increased,SOD decreased) significantly ( all P﹤0. 01)and unsignificantly only in the sham group (all P﹥0. 05); ② compared with ATP group, RES group showed a decreasing in the concentration of MDA, and showed an increasing in the concentration of SOD ( P﹤0. 01 ); compared with RES group and sham group, ATP group showed an increasing in the concentration of MDA, and showed a decreasing in the concentration of SOD ( all P﹤0. 01 ) , there was no difference between the sham group and the RV group in each indicator ( all P ﹥0. 05 ) . Conclusions Oxidative stress reaction was showed markedly in the sustained atrial fibrillation pigs induced by right tachypacing;Resveratrol can significantly inhibit oxidative stress reaction in sustained atrial fibrillation.
