目的探讨阿齐沙坦对转化生长因子β(TGFβ)诱导的自发性高血压大鼠血管平滑肌细胞(VSMCs )增殖及迁移的影响。方法采用组织块贴壁培养法进行血管平滑肌细胞的原代培养,取3~5代细胞用于实验。用MTT法测定细胞增殖情况,用Boyden小室测试细胞迁移情况。采用10ng/ml TGFβ刺激VSMCs ,并用阿齐沙坦干预,24h后用Westernblot和RT PCR检测VSMCs中MMP2、MMP9及TIMP1蛋白和mRNA的表达。结果TGFβ可促进大鼠VSMCs的增殖及迁移,并增加大鼠VSMCs的MMP 2、MMP 9和TIMP 1蛋白表达。阿齐沙坦则可抑制TGFβ诱导的大鼠VSMCs的增殖和迁移,并降低大鼠VSMCs的MMP 2、MMP 9及TIMP 1表达。结论阿齐沙坦可能通过下调MMP 2、MMP 9及TIMP 1的表达来抑制TGFβ诱导的自发性高血压血管平滑肌细胞的增殖及迁移。

목적탐토아제사탄대전화생장인자β(TGFβ)유도적자발성고혈압대서혈관평활기세포(VSMCs )증식급천이적영향。방법채용조직괴첩벽배양법진행혈관평활기세포적원대배양,취3~5대세포용우실험。용MTT법측정세포증식정황,용Boyden소실측시세포천이정황。채용10ng/ml TGFβ자격VSMCs ,병용아제사탄간예,24h후용Westernblot화RT PCR검측VSMCs중MMP2、MMP9급TIMP1단백화mRNA적표체。결과TGFβ가촉진대서VSMCs적증식급천이,병증가대서VSMCs적MMP 2、MMP 9화TIMP 1단백표체。아제사탄칙가억제TGFβ유도적대서VSMCs적증식화천이,병강저대서VSMCs적MMP 2、MMP 9급TIMP 1표체。결론아제사탄가능통과하조MMP 2、MMP 9급TIMP 1적표체래억제TGFβ유도적자발성고혈압혈관평활기세포적증식급천이。
ObjectiveToinvestigatetheeffectsofazilsartanonTGFβ inducedspontaneouslyhypertensivevascularsmoothmuscle cells (VSMCs ) proliferation and migration .Methods Rat VSMCs were cultivated by the method of tissue explants adherence .Cells of generation 3 to 5 were used as the experimental system .The MTT test was used to measure cell proliferation and Boyden chamber as-say was used to measure cell migration .Primary cultured VSMCs were treated by 10 ng/ml TGFβand azilsartan for 24 hours .The expressionofMMP2,MMP9,TIMP1weremeasuredbyWesternblotandRT PCR.ResultsTheexpressionofMMP2,MMP9 and TIMP 1 in rat VSMCs stimulated by TGFβincreased significantly .VSMCs migration and proliferation also increased significantly . Azilsartan significantly inhibited TGFβinduced MMP 2 ,MMP 9 and TIMP 1 production in rat VSMCs ,as well as VSMCs prolifera-tionandmigration.ConclusionsAzilsartaninhibitedTGFβ inducedVSMCsproliferationandmigrationbydown regulationtheex-pression of MMP 2 ,MMP 9 ,TIMP 1 .
