目的:研究趋化因子受体1(CC chemokine receptor 1,CCR1)在人结肠癌组织中的表达分布情况,探讨其表达水平与临床病理特征的关系。方法收集广州军区武汉总医院普通外科手术切除并经病理诊断证实的96例结肠癌组织。采用 Real-time PCR、Western blot及免疫组化技术检测CCR1在96例结肠癌组织及相应的癌旁组织中的表达分布情况,并分析其表达差异与结肠癌淋巴结转移及肝转移的相关性。结果在96例结肠癌组织及其相应的癌旁组织中均可检测到CCR1的表达,结肠癌组织中的表达水平明显高于癌旁组织。CCR1 mRNA 表达水平与患者性别、年龄、肿瘤大小无明显相关性(均P>0.05),而与肿瘤的淋巴结转移、远处转移及TNM分期显著相关(均P<0.05)。免疫组化检测结果显示CCR1在结肠癌组织中的蛋白表达主要定位于细胞膜。26例淋巴转移的结肠癌组织中,10例(38.5%)染色强度为?~?,16例淋巴结转移合并肝转移的结肠癌组织中,13例(81.2%)染色强度为?~?,经χ2检验,两组比较差异具有统计学意义(χ2=6.095,P=0.014)。结论 CCR1在结肠癌组织中的表达高于癌旁组织,其表达水平与淋巴结转移及肝转移相关,具有一定的诊断价值,有望成为判断结肠癌侵袭转移能力及预后的指标。

목적:연구추화인자수체1(CC chemokine receptor 1,CCR1)재인결장암조직중적표체분포정황,탐토기표체수평여림상병리특정적관계。방법수집엄주군구무한총의원보통외과수술절제병경병리진단증실적96례결장암조직。채용 Real-time PCR、Western blot급면역조화기술검측CCR1재96례결장암조직급상응적암방조직중적표체분포정황,병분석기표체차이여결장암림파결전이급간전이적상관성。결과재96례결장암조직급기상응적암방조직중균가검측도CCR1적표체,결장암조직중적표체수평명현고우암방조직。CCR1 mRNA 표체수평여환자성별、년령、종류대소무명현상관성(균P>0.05),이여종류적림파결전이、원처전이급TNM분기현저상관(균P<0.05)。면역조화검측결과현시CCR1재결장암조직중적단백표체주요정위우세포막。26례림파전이적결장암조직중,10례(38.5%)염색강도위?~?,16례림파결전이합병간전이적결장암조직중,13례(81.2%)염색강도위?~?,경χ2검험,량조비교차이구유통계학의의(χ2=6.095,P=0.014)。결론 CCR1재결장암조직중적표체고우암방조직,기표체수평여림파결전이급간전이상관,구유일정적진단개치,유망성위판단결장암침습전이능력급예후적지표。
Objective To investigate the expression of CC chemokine receptor 1(CCR1)in the human colorectal carcinoma and its correlation with clinicopathological characteristics of colorectal carcinoma.Methods A total of 9 6 colorectal carcinoma tissues and their adjacent normal tissues were surgically removed and pathologically confirmed in our hospital.Real-time PCR, Western blot and immunohistochemistry were used to examine the expression and distribution of CCR1 in these tissues.Moreover,the association of the expression of CCR1 with lymph nodes metastasis and liver metastasis was ana-lyzed.Results CCR1 was found to express at mRNA and protein levels in 96 colorectal carcinoma tissues and their adjacent normal tissues,with the expression level significantly higher in cancer tissues than in adjacent normal tissues(P<0.05).The expression of CCR1 mRNA was significantly correlated with lymph nodes metastasis,remote metastasis and TNM stage(P<0.05 for all),but not with gender,age and tumor size.In addition,immunohistochemistry revealed that CCR1 protein was main-ly located on the membrane of colorectal carcinoma cells.Moreover,the moderate to strong staining of CCR1 protein was found in 81.2% of colorectal carcinomas with lymph nodes metastasis and liver metastasis,which was markedly higher than in 38.5%of colorectal carcinomas with only lymph nodes metastasis(χ2=6.095,P=0.014).Conclusion The expression of CCR1 is greater in colorectal cancer than in adjacent normal tissues.CCR1 expression levels are correlated with lymph nodes metastasis and liver metastasis,which is of clinical value.CCR1 may serve as a potential marker to evaluate the metastasis of colorectal carcinoma and the prognosis of patients.
