目的:探讨黄芪皂苷对慢性心衰大鼠心室重塑及血浆脑钠肽水平的影响。方法利用结扎左冠状动脉前降支复制心梗后心衰大鼠模型,造模成功后随机分为模型组、黄芪皂苷低、高剂量组(10、20 mg/kg)。假手术组仅冠状动脉前降支下穿线不结扎。每组15只。术后第2天开始灌胃给药,假手术组和模型组给予等体积生理盐水灌胃,每天1次。连续给药4周后采用心脏超声检测各组大鼠左室舒张末期内径( LVEDd)、左室收缩末期内径( LVEDs)、左室射血分数( LVEF)、左、右心室质量指数,检测血浆BNP水平。结果与假手术组比较,模型组 LVEDd、LVEDs 显著增加(P<0.05),LVEF 明显下降(P<0.05),左、右心室质量指数明显增加(P<0.01),血浆BNP 表达增加(P<0.05)。与模型组比较,黄芪皂苷低、高剂量组大鼠LVEDd、LVEDs减少(P<0.05),LVEF增加(P<0.05);左、右心室质量指数降低(P<0.01);血浆BNP水平降低( P<0.05)。结论黄芪皂苷可以降低血浆BNP水平,延缓心衰大鼠模型的心室重塑,改善心功能。

목적:탐토황기조감대만성심쇠대서심실중소급혈장뇌납태수평적영향。방법이용결찰좌관상동맥전강지복제심경후심쇠대서모형,조모성공후수궤분위모형조、황기조감저、고제량조(10、20 mg/kg)。가수술조부관상동맥전강지하천선불결찰。매조15지。술후제2천개시관위급약,가수술조화모형조급여등체적생리염수관위,매천1차。련속급약4주후채용심장초성검측각조대서좌실서장말기내경( LVEDd)、좌실수축말기내경( LVEDs)、좌실사혈분수( LVEF)、좌、우심실질량지수,검측혈장BNP수평。결과여가수술조비교,모형조 LVEDd、LVEDs 현저증가(P<0.05),LVEF 명현하강(P<0.05),좌、우심실질량지수명현증가(P<0.01),혈장BNP 표체증가(P<0.05)。여모형조비교,황기조감저、고제량조대서LVEDd、LVEDs감소(P<0.05),LVEF증가(P<0.05);좌、우심실질량지수강저(P<0.01);혈장BNP수평강저( P<0.05)。결론황기조감가이강저혈장BNP수평,연완심쇠대서모형적심실중소,개선심공능。
Objective To observe the effect of astragaloside( AST) on the ventricular remodeling and plasma brain natriuretic peptide(BNP) expression in rats with chronic heart failure. Methods The animal model of myocardial infarction was established by ligation of the left anterior descending coronary artery. Animals were randomly divided into 4 groups,including the model group,two different doses of AST groups( 10 and 20 mg/kg) and the sham operation group. Intragastric administration of AST or physiological saline began day 2 after operation. After four weeks,LVEDd,LVEDs,LVEF and left/right ventricular mass index were detected by cardiac ultrasound. The level of plasma BNP was measured. Results Compared with the sham operation group,LVEDd and LVEDs were increased obviously(P<0.05),while LVEF was significantly decreased( P<0.05) in the model group. Meantime,left/right ventricular mass index and plasma BNP level were increased significantly in the model group. Compared with the model group,LVEDd and LVEDs were significantly decreased( P<0.05) ,but LVEDs was increased obviously( P<0.05) in the AST-treated group. Meantime,left/right ventricular mass index and plasma BNP level were decreased significantly. Conclusion AST can reduce BNP level in plasma,delay myocardial remodeling in rats with myocardial infarction and improve cardiac function.
