目的:通过观察云南灯盏花胶囊对5/6肾切除慢性肾衰竭大鼠结缔组织生长因子(connectivetissuegrowthfactor,CTGF)、Smad7蛋白表达的影响,探讨云南灯盏花胶囊减轻肾纤维化以延缓慢性肾衰竭的作用机制。方法采用5/6肾切除法造模,观察实验大鼠治疗后的一般状态。检测血肌酐(serumcreatinine,Scr)、血尿素氮(bloodureanitrogen,BUN)摘取残肾组织,RT-PCR检测肾组织CTGF的表达,蛋白质印迹技术(WesternBlot)法检测肾组织内Smad7mRNA的表达。结果云南灯盏花胶囊组慢性肾衰竭(chronicrenalfailure,CRF)大鼠BUN、Scr下降,肾组织CTGF表达减弱,Smad7mRNA表达增强,与模型组比较差异有统计学意义(P<0.05)。结论云南灯盏花胶囊可明显改善5/6肾切除CRF大鼠肾功能,升高Smad7mRNA表达,减少肾纤维化因子CTGF表达,延缓慢性肾衰竭进展。

목적:통과관찰운남등잔화효낭대5/6신절제만성신쇠갈대서결체조직생장인자(connectivetissuegrowthfactor,CTGF)、Smad7단백표체적영향,탐토운남등잔화효낭감경신섬유화이연완만성신쇠갈적작용궤제。방법채용5/6신절제법조모,관찰실험대서치료후적일반상태。검측혈기항(serumcreatinine,Scr)、혈뇨소담(bloodureanitrogen,BUN)적취잔신조직,RT-PCR검측신조직CTGF적표체,단백질인적기술(WesternBlot)법검측신조직내Smad7mRNA적표체。결과운남등잔화효낭조만성신쇠갈(chronicrenalfailure,CRF)대서BUN、Scr하강,신조직CTGF표체감약,Smad7mRNA표체증강,여모형조비교차이유통계학의의(P<0.05)。결론운남등잔화효낭가명현개선5/6신절제CRF대서신공능,승고Smad7mRNA표체,감소신섬유화인자CTGF표체,연완만성신쇠갈진전。
Objective By observing the Yunnan Breviscapine capsules on 5/6 nephrectomy chron-ic renal failure rats the influence of connective tissue growth factor ( CTGF) ,Smad7 protein expression , dis-cuss the mechanism of Yunnan Breviscapine capsules on relieving renal fibrosis to delay the chronic renal failure.Methods 5/6 renal resection method is adopted to build mould , observe the experimental rats'general status after treatment .Serum creatinine , and blood urea nitrogen are detected , and residual kidney tissues are picked , RT-PCR is used to detect renal tissue CTGF expression , and Protein imprinting technol-ogy(Western Blot) is used to detect renal tissues Smad7 mRNA expression.Results The BUN and Scr Decreased in Yunnan Breviscapine capsules group of chronic renal failure rats .CTGF express abate of Kid-ney tissue,Smad7 mRNA Express to enhance of Kidney tissue , Compared with model group , difference was statistically significant(P<0.05).Conclusion Yunnan Breviscapine capsules can obviously improve the 5/6 nephrectomy CRF rats'renal function , elevate Smad7 mRNA expression , reduce renal fibrosis factor CTGF expression , and delay chronic renal failure .
