目的:评价两种方法对蒙药金诃子低温提取物多糖成分和含量的检测效果。方法采用红外光谱技术和硫酸-蒽酮比色法测定蒙药金诃子5个不同部位提取物中多糖的成分和含量,红外光谱技术检测部分由清华大学分析中心完成。评价硫酸-蒽酮比色法的精密度、稳定性、重复性和加样回收率等性能指标。结果红外光谱技术测得蒙药金诃子5个部位提取物中均有多糖成分,但各部位的含量比较差异无统计学意义。硫酸-蒽酮比色法线性范围为40~200μg/mL;精密度良好( RSD=0.460%),重复性良好( RSD=1.616%),平均加标回收率为(110.130±0.010)%( RSD=0.946%);试剂在室温自然光下2 h内稳定性良好;标准曲线线性回归方程为Y=0.2051X-0.0034,R=0.9991;蒙药金诃子5个部位提取物中多糖含量为14.15%~22.15%。结论蒙药金诃子不同部位提取物的成分分析优选红外光谱技术,而硫酸-蒽酮比色法用于检测多糖含量。

Objective To estimate the detection results of the two methods on the determination of components and con -tent of polysaccharides in low temperature extracts from Mongolian Terminalia chebula Ritz .Methods The components and content of polysaccharides in five different positions were determined by infrared spectroscopy and anthrone -sulfuric acid color-imetric method.The infrared spectroscopy detection was completed in Tsinghua university analysis center .The precision, sta-bility, repeatability and average recovery rate of anthrone-sulfuric acid colorimetric method were evaluated by different experi-ments.Results The polysaccharides in the five different positions were determined by infrared spectroscopy , but no signifi-cant difference was found among the five different positions .In anthrone-sulfuric acid colorimetric method , the linear ranges of polysaccharides were 40-200 μg/mL, the precision was good ( RSD =0.460%) , the repeatability was good ( RSD =1.616%), and the average recovery rate was (110.130 ±0.010)%(RSD=0.946%).The reagent stability was good in 2 h at room temperature under natural light; linear regression equation of standard curve was Y =0.205 1X-0.003 4, R=0.999 1.There were polysaccharides in the five different positions , and the contents of polysaccharides were 14.15%-22.15%.Conclusions The component analysis of polysaccharides in the five different positions is preferably detected by in -frared spectroscopy , and anthrone-sulfuric acid colorimetric method is used to determine the contents of polysaccharides .
