目的:鉴定核桃楸皮的化学成分,并对其进行体外抗肝癌活性成分筛选。方法采用75%乙醇回流法提取核桃楸皮,经分级萃取、硅胶柱层析及制备型高效液相色谱分析等方法得到单体化合物,通过化合物的理化性质及核磁共振技术鉴定化合物成分;采用MTT法评价其体外抗肝癌活性。结果从核桃楸皮中分离得到2种黄酮类化合物,经鉴定分别为木犀草素和刺芒柄花素。 MTT检测结果表明,木犀草素对肝癌细胞HepG2和正常肝细胞LO2都有一定的毒性作用,IC50分别为(103.32±21.34)、(131.13±13.48)μmol/L;刺芒柄花素对肝癌细胞HepG2的毒性作用较小,其IC50>200μmol/L。结论核桃楸皮含2种黄酮类化合物:木犀草素和刺芒柄花素,其中木犀草素具有明显的抗肝癌作用。

목적:감정핵도추피적화학성분,병대기진행체외항간암활성성분사선。방법채용75%을순회류법제취핵도추피,경분급췌취、규효주층석급제비형고효액상색보분석등방법득도단체화합물,통과화합물적이화성질급핵자공진기술감정화합물성분;채용MTT법평개기체외항간암활성。결과종핵도추피중분리득도2충황동류화합물,경감정분별위목서초소화자망병화소。 MTT검측결과표명,목서초소대간암세포HepG2화정상간세포LO2도유일정적독성작용,IC50분별위(103.32±21.34)、(131.13±13.48)μmol/L;자망병화소대간암세포HepG2적독성작용교소,기IC50>200μmol/L。결론핵도추피함2충황동류화합물:목서초소화자망병화소,기중목서초소구유명현적항간암작용。
Objective To investigate the chemical compositions of Cortex Juglans Mandshurica and to evaluate anti-tumor effects of these compounds in human hepatoma cells.Methods Cortex Juglans Mandshurica were extracted with 75%ethanol, concentrated with chloroform and ethyl acetate, eluted radiantly on silica gel, and purified by preparative HPLC.Physicochemi-cal constants and spectroscopic analysis were used to identify their structures .MTT assay was used to evaluate their anti-hepato-carcinoma bioactivities.Results Two compounds were isolated and elucidated as luteolin and formononetin.Luteolin had strong cytotoxic effect on HepG2 and LO2 with IC50 values of (103.32 ±21.34) and (131.13 ±13.48) μmol/L;Formononetin showed lighter cytotoxic effect on HepG2 with IC50>200μmol/L.Conclusion Luteolin and formononetin are isolated from Cortex Jug-lans Mandshurica for the first time, and luteolin shows significant anti-tumor activity in human hepatoma cells.
