目的:研究大鼠灌胃缩泉胶囊后,益智黄酮(杨芽黄素、伊砂黄素、白杨素、山奈素和芹菜素-4',7-二甲氧醚)二相结合产物的酶水解。方法雄性Sprague-Dawley大鼠胆管插管手术完毕后经灌胃给予缩泉胶囊(1.64 g/kg)后收集0~4 h胆汁。胆汁样品经β-葡萄糖醛酸水解酶(1000 U/ml,pH为5的乙酸钠缓冲液)处理后,利用液相色谱-质谱联用仪(LC-MS/MS)进行分析。结果益智黄酮的葡萄糖醛酸结合物在酶水解处理后,代谢物的色谱峰消失,释放出黄酮的原形物质形式。虽然脱甲基反应后形成葡萄糖醛酸结合物也被水解,但并未能检测到脱甲基产物。结论β-葡萄糖醛酸水解酶处理可进一步确定益智黄酮在大鼠胆汁中的物质形式,也为定量分析奠定基础。

목적:연구대서관위축천효낭후,익지황동(양아황소、이사황소、백양소、산내소화근채소-4',7-이갑양미)이상결합산물적매수해。방법웅성Sprague-Dawley대서담관삽관수술완필후경관위급여축천효낭(1.64 g/kg)후수집0~4 h담즙。담즙양품경β-포도당철산수해매(1000 U/ml,pH위5적을산납완충액)처리후,이용액상색보-질보련용의(LC-MS/MS)진행분석。결과익지황동적포도당철산결합물재매수해처리후,대사물적색보봉소실,석방출황동적원형물질형식。수연탈갑기반응후형성포도당철산결합물야피수해,단병미능검측도탈갑기산물。결론β-포도당철산수해매처리가진일보학정익지황동재대서담즙중적물질형식,야위정량분석전정기출。
Objective To study enzymatic hydrolysis of the flavonoids' (tectochrysin, izalpinin, chrysin, kaempferide and apigenin-4',7-dimethylether) metabolites in rat bile after oral administration of SuoQuan cap-sules. Methods Cannulas were surgically inserted into the bile duct of male Sprague-Dawley rats (n=3) under an-esthesia (pentobarbital, 50 mg/kg, i.p.), and the bile was collected for 4 h after oral administration of Suoquan cap-sules (1.64 g/kg, suspended in 0.5%w/v sodium carboxymethycellulose). Afterβ-glucuronidase (1000 units/ml in pH 5 acetate buffer) treatment, these phaseⅡmetabolites in rat bile samples were hydrolyzed and their aglycones were analyzed using liquid chromatography/tandem mass spectrometry (LC-MS/MS) with selected reaction monitoring mode. Results After β-glucuronidase treatment, the mono-or di-glucuronide metabolites of Yizhi flavonoids (tec-tochrysin, izalpinin, chrysin, kaempferide and apigenin-4', 7-dimethylether) could not be detected. On the contrast, the corresponding aglycones were librated from their conjugated forms. However, the O-demethylation metabolites of izalpinin and apigenin-4', 7-dimethylether could not be measured, although their glucuronidated conjugates had been found in the rat bile samples. Conclusion The enzymatic hydrolysis viaβ-glucuronidase is helpful for the identifica-tion of Yizhi flavonoids' phase Ⅱ metabolites. Meanwhile, this paper also lays a partial foundation for quantitative analysis of these aglycones.
