目的:研究白藜芦醇对TRAIL诱导肝癌细胞SMMC-7721凋亡的影响及其机制。方法培养人肝癌细胞株SMMC-7721,分为对照组、TRAIL干预组、Res +TRAIL干预组,进行DNA末端原位标记( TUNEL )染色法作凋亡分析;Western Blot 检测白藜芦醇对SMMC-7721细胞 Survivn 表达的影响。结果对照组SMMC -7721细胞的凋亡指数为(1.78±04.9)%,TRAIL 干预组凋亡指数为(9.86±0.53)%,Res+TRAIL干预组中25μmol/L、50μmol/L、100μmol/L浓度Ros凋亡指数分别为(11.30±0.80)%,(20.89±0.69)%和(27.78±1.88)%。除Res +TRAIL干预组(25μmol/L)与TRAIL干预组比较无显著性差异(P>0.05),其余各组之间比较差异均有统计学意义(P均<0.05)。 Western Blot测定结果显示:25μmol/L Ros处理组Survivin /β-actin的值与对照组相比无统计学差异(P>0.05);50μmol/L和100μmol/L Res处理组Survivin /β-actin的值较对照组显著降低(P<0.01);100μmol/L Res处理组Survivin /β-actin的值较50μmol/L显著降低(P<0.01)。结论 TRAIL对SMMC-7721细胞有凋亡诱导作用;Res对TRAIL诱导人肝癌细胞SMMC-7721凋亡有致敏作用,并且这种作用呈剂量依赖关系;其分子机制可能与下调凋亡抑制因子Survivin的表达有关。

목적:연구백려호순대TRAIL유도간암세포SMMC-7721조망적영향급기궤제。방법배양인간암세포주SMMC-7721,분위대조조、TRAIL간예조、Res +TRAIL간예조,진행DNA말단원위표기( TUNEL )염색법작조망분석;Western Blot 검측백려호순대SMMC-7721세포 Survivn 표체적영향。결과대조조SMMC -7721세포적조망지수위(1.78±04.9)%,TRAIL 간예조조망지수위(9.86±0.53)%,Res+TRAIL간예조중25μmol/L、50μmol/L、100μmol/L농도Ros조망지수분별위(11.30±0.80)%,(20.89±0.69)%화(27.78±1.88)%。제Res +TRAIL간예조(25μmol/L)여TRAIL간예조비교무현저성차이(P>0.05),기여각조지간비교차이균유통계학의의(P균<0.05)。 Western Blot측정결과현시:25μmol/L Ros처리조Survivin /β-actin적치여대조조상비무통계학차이(P>0.05);50μmol/L화100μmol/L Res처리조Survivin /β-actin적치교대조조현저강저(P<0.01);100μmol/L Res처리조Survivin /β-actin적치교50μmol/L현저강저(P<0.01)。결론 TRAIL대SMMC-7721세포유조망유도작용;Res대TRAIL유도인간암세포SMMC-7721조망유치민작용,병차저충작용정제량의뢰관계;기분자궤제가능여하조조망억제인자Survivin적표체유관。
Objective It is to observe the influence of resveratrol on TRAIL -induced apoptosis in human hepatoma cell line SMMC-7721 and its mechanism .Methods SMMC-7721 cells were cultured in vitro and divided into three gropus:con-trol group, TRAIL intervention group, resveratrol and TRAIL intervention group .After treatment with TRAIL/resveratrol and TRAIL for 24 hours, TUNEL staining were given to detect the effects of resveratrol on TRAIL -induced apoptosis in human hepatoma cell line SMMC -7721;Western blot technology was performed to detect the expression of apoptosis -related protein survivin in hepatoma apoptotic cells treated by resveratrol .Results The apoptotic rate of SMMC-7721 cells was (1.78 ±0. 49)%in control group, and was (9.86 ±0.53)% in TRAIL intervention group, was (11.30 ±0.80)%, (20.89 ±0. 69)%, (27.78 ±1.88)%respectively after cotreatment with 100 μg/L TRAIL and 25 μmol/L or 50 μmol/L or 100 μmol/L resveratrol for 24 hours in resveratrol and TRAIL intervention group .There was significant difference between every two groups, excepted 25 μmol/L resveratrol intervention group with TRAIL intervention group (P<0.05).The results of detec-tion with Western Blot technology indicated that after being treated by 50 μmol/L, 100 μmol/L resveratrol for 24 hours, the value of surviving/β-actin was down-regulated significantly compared with control group (P<0.01);and there was signifi-cant difference between 50 μmol/L and 100 μmol/L resveratrol-treated group (P<0.01).Conclusion TRAIL can induce apoptosis of human hepatoma SMMC -7721 cells in vitro.Resveratrol can enhance TRAIL -induced apoptosis of SMMC -7721 cells in dose-dependent manner .This enhancement may be related to the inhibition of expression of survivin by resveratrol .
