目的:探讨葡萄籽原花青素对大鼠肝缺血再灌注损伤的抗氧化作用、凋亡相关蛋白Bcl-2和Bax表达的影响。方法:SD雄性大鼠30只,复制肝缺血再灌注损伤模型。随机分为:假手术组;HIRI模型组;葡萄籽原花青素预处理(GSPE)组(160 mg/kg),每组10只。于再灌注后3 h检测肝组织谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、丙二醛(MDA)含量及血清谷丙转氨酶(ALT)、谷草转氨酶(AST)的含量;光镜下比较各组肝组织形态学的变化;免疫组化法检测GSPE对凋亡相关蛋白Bcl-2,Bax表达的影响。结果:与HIRI模型组比较,GSPE预处理组血清ALT、AST水平明显降低(P<0.05);肝组织GSH-Px、SOD的活性明显升高(P<0.05),而MDA含量下降(P<0.05)。光镜下GSPE预处理组肝细胞损伤程度较模型组明显减轻;免疫组化结果显示,GSPE预处理组肝组织Bcl-2蛋白的表达量较模型组明显增多,而Bax蛋白的表达量则较模型组明显减少。结论:GSPE对缺血再灌注肝脏具有一定的抗氧化损伤和抗凋亡的作用。

목적:탐토포도자원화청소대대서간결혈재관주손상적항양화작용、조망상관단백Bcl-2화Bax표체적영향。방법:SD웅성대서30지,복제간결혈재관주손상모형。수궤분위:가수술조;HIRI모형조;포도자원화청소예처리(GSPE)조(160 mg/kg),매조10지。우재관주후3 h검측간조직곡광감태과양화물매(GSH-Px)、초양화물기화매(SOD)、병이철(MDA)함량급혈청곡병전안매(ALT)、곡초전안매(AST)적함량;광경하비교각조간조직형태학적변화;면역조화법검측GSPE대조망상관단백Bcl-2,Bax표체적영향。결과:여HIRI모형조비교,GSPE예처리조혈청ALT、AST수평명현강저(P<0.05);간조직GSH-Px、SOD적활성명현승고(P<0.05),이MDA함량하강(P<0.05)。광경하GSPE예처리조간세포손상정도교모형조명현감경;면역조화결과현시,GSPE예처리조간조직Bcl-2단백적표체량교모형조명현증다,이Bax단백적표체량칙교모형조명현감소。결론:GSPE대결혈재관주간장구유일정적항양화손상화항조망적작용。
Objective: To study the effects of grape seed proanthocyanidin extract (GSPE) on antioxidation of hepatic ischemia reperfusion injury in rats, and the expression of apoptosis-related proteins Bcl-2 and Bax. Methods: Thirty SD male rats were randomly divided into three groups with 10 rats each .i.e. sham operation group, HIRI model group and GSPE preconditioning group (160 mg/kg). The content of GSH-Px, SOD, and MDA in hepatic tissues, and the content of ALT and AST in serum were assayed 3 hours later after reperfusion. The morphological variations of hepatic tissues in each group were compared by means of optical microscope. Effects of GSPE on Bcl-2 and Bax in hepatic tissues were tested by means of immunohistochemisty. Results:The content of ALT and AST in serum and MDA in hepatic tissues decreased significantly(P<0.05), and the content of GSH-Px and SOD in hepatic tissues increased remarkably(P<0.05)in GSPE preconditioning group, compared with HIRI model;Observed by the optical microscope, the degree of liver cell injury in GSPE preconditioning group was remarkably slighter than that of HIRI model group. Immunohistochemical results showed that the content of Bcl-2 increased and Bax decreased significantly in hepatic tissues, compared with that in HIRI model(P<0.05). Conclusion: GSPE had some effects on antioxidation and anti-apoptosis of hepatic ischemia reperfusion injury.
