观察三痹颗粒对Ⅱ型胶原诱发类风湿关节炎(CIA)模型大鼠病变关节细胞趋化因子受体的影响,探讨其抗炎作用机制。方法:将 CIA 模型大鼠随机分为模型组、三痹颗粒高剂量组、三痹颗粒中剂量组、三痹颗粒低剂量组、另取空白组。三痹颗粒高、中、低剂量组于初次免疫第15天分别给予三痹颗粒蒸馏水溶液灌胃,模型组和空白对照组灌服同体积的生理盐水,连续灌服20天,观察大鼠足踝关节肿胀度以及关节炎指数的变化,RT -PCR 法测定大鼠外周血趋化因子受体3(CXCR3)、趋化因子受体4(CXCR4)mRNA 含量。结果:第21天,与空白组比较,模型组及三痹颗粒各组足容积明显增大,差异有统计学意义(P <0.05);第35天时,与模型组比较,三痹颗粒高、中剂量组足容积的上升程度减慢,进一步减轻关节炎症反应程度,差异有统计学意义(P <0.05)。与模型组比较,中药各剂量组大鼠关节炎评分指数明显降低,差异有统计学意义( P <0.05)。与模型组比较,三痹颗粒各组大鼠外周血CXCR3mRNA、CXCR4mRNA 的表达明显降低,差异有统计学意义(P <0.05)。结论:三痹颗粒能够抑制CIA 模型大鼠外周血 CXCR3、CXCR4的表达,从而减慢 CIA 模型大鼠足容积的上升程度,降低关节炎指数,抑制炎症反应。
觀察三痺顆粒對Ⅱ型膠原誘髮類風濕關節炎(CIA)模型大鼠病變關節細胞趨化因子受體的影響,探討其抗炎作用機製。方法:將 CIA 模型大鼠隨機分為模型組、三痺顆粒高劑量組、三痺顆粒中劑量組、三痺顆粒低劑量組、另取空白組。三痺顆粒高、中、低劑量組于初次免疫第15天分彆給予三痺顆粒蒸餾水溶液灌胃,模型組和空白對照組灌服同體積的生理鹽水,連續灌服20天,觀察大鼠足踝關節腫脹度以及關節炎指數的變化,RT -PCR 法測定大鼠外週血趨化因子受體3(CXCR3)、趨化因子受體4(CXCR4)mRNA 含量。結果:第21天,與空白組比較,模型組及三痺顆粒各組足容積明顯增大,差異有統計學意義(P <0.05);第35天時,與模型組比較,三痺顆粒高、中劑量組足容積的上升程度減慢,進一步減輕關節炎癥反應程度,差異有統計學意義(P <0.05)。與模型組比較,中藥各劑量組大鼠關節炎評分指數明顯降低,差異有統計學意義( P <0.05)。與模型組比較,三痺顆粒各組大鼠外週血CXCR3mRNA、CXCR4mRNA 的錶達明顯降低,差異有統計學意義(P <0.05)。結論:三痺顆粒能夠抑製CIA 模型大鼠外週血 CXCR3、CXCR4的錶達,從而減慢 CIA 模型大鼠足容積的上升程度,降低關節炎指數,抑製炎癥反應。
관찰삼비과립대Ⅱ형효원유발류풍습관절염(CIA)모형대서병변관절세포추화인자수체적영향,탐토기항염작용궤제。방법:장 CIA 모형대서수궤분위모형조、삼비과립고제량조、삼비과립중제량조、삼비과립저제량조、령취공백조。삼비과립고、중、저제량조우초차면역제15천분별급여삼비과립증류수용액관위,모형조화공백대조조관복동체적적생리염수,련속관복20천,관찰대서족과관절종창도이급관절염지수적변화,RT -PCR 법측정대서외주혈추화인자수체3(CXCR3)、추화인자수체4(CXCR4)mRNA 함량。결과:제21천,여공백조비교,모형조급삼비과립각조족용적명현증대,차이유통계학의의(P <0.05);제35천시,여모형조비교,삼비과립고、중제량조족용적적상승정도감만,진일보감경관절염증반응정도,차이유통계학의의(P <0.05)。여모형조비교,중약각제량조대서관절염평분지수명현강저,차이유통계학의의( P <0.05)。여모형조비교,삼비과립각조대서외주혈CXCR3mRNA、CXCR4mRNA 적표체명현강저,차이유통계학의의(P <0.05)。결론:삼비과립능구억제CIA 모형대서외주혈 CXCR3、CXCR4적표체,종이감만 CIA 모형대서족용적적상승정도,강저관절염지수,억제염증반응。
Objective:To observe the effect of Sanbi granule on chemokine receptor of diseased joints in CIA model rats , and to discuss the anti -inflammatory mechanism .Methods:Divide CIA model rats randomly into a model group ,a high dose group ,a mid-dose group ,a low-dose group and a blank group .In high dose group ,mid-dose group and low -dose group model rats were given different doses of Sanbi granule distilled solution in primary immune on the 15th day. In model group and blank group model rats were given the same volume normal saline .All model rats were gavaged for 20 days.Observe model rats ’ swelling ankles and the arthritis index .Measure the contents of CXCR 3 and CXCR4 in model rats with RT -PCR method.Results:Compared with blank group on the 2lst day, in model group and Sanbi granule groups the model rats ’ feet were all increased in volume .The difference had a statistical significance ( P<0 .05 ) .Com-pared with model group on the 35th day,the rising degree of model rats ’ feet volume in Sanbi granule groups slowed down .And in these groups the degree of joint inflammation was reduced .The difference had a statistical significance ( P<0.05 ) .Compared with model group ,the arthritis index of Sanbi granule groups was decreased sharply .The differ-ence had a statistical significance (P <0.05).Compared with model group,the expressions of CXCR3mRNA,CX-CR4mRNA of model rats ’ peripheral blood in Sanbi granule groups were decreased obviously .The difference had a sta-tistical significance(P<0.05).Conclusion:Sanbi granule can restrain the expressions of CXCR 3,CXCR4 of peripheral blood in CIA model rats ,make the rising degree of model rats ’ feet volume to slow down ,decrease arthritis index and re-strain inflammatory reaction .
