目的:探讨择时电针治疗氯胺酮滥用成瘾的某些神经生物学机制。方法将56只清洁级 SD 大鼠随机分为7组,即正常组、生理盐水组、模型组、子时电针组、卯时电针组、午时电针组和酉时电针组,每组8只。每天1次经腹腔注射盐酸氯胺酮注射液100 mg/kg ,连续给药7 d 复制氯胺酮滥用成瘾模型,不同时辰电针组在造模成功后选取一侧“三阴交”和“足三里”穴给予电针(低频2 Hz)治疗,每次30 min ,连续7 d 。采用免疫组织化学染色方法检测隔内侧核(medial septal nucleus , MS)酪氨酸羟化酶(tyrosine hydroxylase ,TH)、c-fos的表达。结果与正常组和生理盐水组相比较,模型组MSTH、c-fos表达明显增强(P<0.05)。与模型组相比较,午时、酉时电针组MSTH、c-fos表达明显减弱(P<0.05),而子时、卯时电针组无明显变化(P>0.05)。结论氯胺酮滥用成瘾可以增强TH、c-fos在MS的表达,午时、酉时电针具有逆转作用。

목적:탐토택시전침치료록알동람용성은적모사신경생물학궤제。방법장56지청길급 SD 대서수궤분위7조,즉정상조、생리염수조、모형조、자시전침조、묘시전침조、오시전침조화유시전침조,매조8지。매천1차경복강주사염산록알동주사액100 mg/kg ,련속급약7 d 복제록알동람용성은모형,불동시신전침조재조모성공후선취일측“삼음교”화“족삼리”혈급여전침(저빈2 Hz)치료,매차30 min ,련속7 d 。채용면역조직화학염색방법검측격내측핵(medial septal nucleus , MS)락안산간화매(tyrosine hydroxylase ,TH)、c-fos적표체。결과여정상조화생리염수조상비교,모형조MSTH、c-fos표체명현증강(P<0.05)。여모형조상비교,오시、유시전침조MSTH、c-fos표체명현감약(P<0.05),이자시、묘시전침조무명현변화(P>0.05)。결론록알동람용성은가이증강TH、c-fos재MS적표체,오시、유시전침구유역전작용。
Objective To explore some of the neurobiological mechanisms involved in the effect of electroacupuncture at different times on ketamine abuse addiction .Methods A total of 56 Sprague-Dawley rats were randomized into normal control group ,normal saline group(2 ml/kg) ,model group ,and Zi-Shi (23 :00-1 :00) EA group ,Mao-Shi (5 :00-7 :00) EA group ,Wu-Shi (11 :00-13 :00) EA group and You-Shi (17 :00-19 :00) EA group .Ketamine abuse model was established by intraperitoneal administration of ket-amine (100 mg /kg) ,once a day for 7 days .EA (2Hz ,3v) was applied to unilaterd “Sanyinjiao” (SP6) and “Zusanli” (ST36) for 30 min ,once daily for 7 days .The expression of tyrosine hydroxylase (TH) and c-fos in the MS was detected by immunohistochemistry .Results Compared with that of the normal control group ,TH and c-fos expression increased in MS of the ketamine group (P< 0 .05) .Compared with that of the model group ,Wu-Shi and You-Shi EA groups reduced TH and c-fos expression (P< 0 .05) ,while Zi-Shi and Mao-Shi EA groups had no obvious effect (P > 0 .05) .Conclusion Ketamine abuse addiction can promote TH and c-fos expression in the medial septal nucleus ,and Wu-Shi and You-Shi electroacupunc-ture can reverse this effect .
