目的:探讨慢性束缚应激对大鼠在旷场迷宫中的行为学参数和大脑杏仁核部位5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)含量的影响,以及加味甘麦大枣汤的干预作用。方法:采用经典的慢性束缚应激的实验方法,复制大鼠慢性应激的模型,然后记录大鼠在旷场迷宫中的行为学参数;分离大鼠大脑杏仁核部位,采用酶联免疫吸附法测定其中5-HT、5-HIAA 的浓度,进而推算出慢性束缚应激对杏仁核部位5-HIAA/5-HT 比值的影响,并探究加味甘麦大枣汤对其的干预作用。结果:实验结果表明,与空白组相比,模型组大鼠在穿格数、直立次数和理毛次数这三个行为学参数上明显减少(P <0.05),而排便次数这个参数则显著增加(P <0.05);杏仁核部位5-HT、5-HIAA 含量以及5-HIAA/5-HT 比值明显减小(P <0.05)。与模型组相比,给药组大鼠在穿格数和理毛次数这两个指标上显著增加(P <0.05),排便次数明显减少(P <0.05);在直立次数这个指标上虽有增加的趋势,但无统计学意义(P >0.05)。杏仁核部位5-HT、5-HIAA 的含量显著升高(P <0.05),虽然5-HIAA/5-HT 的比值有升高的趋势,但是无统计学上的差异(P >0.05)。结论:慢性束缚应激使大鼠在旷场迷宫中的各行为学参数明显改变,使大鼠杏仁核部位5-HT 和其代谢产物的含量以及5-HT 能神经系统的兴奋性降低。中药复方加味甘麦大枣汤能明显对抗慢性束缚应激对大鼠造成的影响。

목적:탐토만성속박응격대대서재광장미궁중적행위학삼수화대뇌행인핵부위5-간색알(5-HT)、5-간신타을산(5-HIAA)함량적영향,이급가미감맥대조탕적간예작용。방법:채용경전적만성속박응격적실험방법,복제대서만성응격적모형,연후기록대서재광장미궁중적행위학삼수;분리대서대뇌행인핵부위,채용매련면역흡부법측정기중5-HT、5-HIAA 적농도,진이추산출만성속박응격대행인핵부위5-HIAA/5-HT 비치적영향,병탐구가미감맥대조탕대기적간예작용。결과:실험결과표명,여공백조상비,모형조대서재천격수、직립차수화리모차수저삼개행위학삼수상명현감소(P <0.05),이배편차수저개삼수칙현저증가(P <0.05);행인핵부위5-HT、5-HIAA 함량이급5-HIAA/5-HT 비치명현감소(P <0.05)。여모형조상비,급약조대서재천격수화리모차수저량개지표상현저증가(P <0.05),배편차수명현감소(P <0.05);재직립차수저개지표상수유증가적추세,단무통계학의의(P >0.05)。행인핵부위5-HT、5-HIAA 적함량현저승고(P <0.05),수연5-HIAA/5-HT 적비치유승고적추세,단시무통계학상적차이(P >0.05)。결론:만성속박응격사대서재광장미궁중적각행위학삼수명현개변,사대서행인핵부위5-HT 화기대사산물적함량이급5-HT 능신경계통적흥강성강저。중약복방가미감맥대조탕능명현대항만성속박응격대대서조성적영향。
Objective:To investigate the effect of chronic restraint stress on rats ’ behavioral parameters in open field maze and the levels of 5 -HT, 5 -HIAA in amygdala, and the intervention of Jiawei Ganmai Dazao Tang (JWGMDZT). Methods: In this research, chronic restraint stress method was used to establish the model of chronic mild stress in rats . Behavioral parameters of rats in open field maze and the levels of 5 -HT, 5 -HIAA, 5 -HIAA/5 -HT in amygdala were evaluaed.The intervention of JWGMDZT was also investigated .Results: Compared with vehicle, the number of crossing line, rearing and grooming in open field maze significantly reduced (P <0.05), while defaecation increased markedly(P <0.05).The levels of 5 -HT, 5 -HIAA and 5 -HIAA/5 -HT in amygdala reduced significantly (P <0.05).Compared with model group, the number of crossing line and grooming significantly increased (P <0.05), while defaecation reduced markedly(P <0.05).Though there was an increased trend of the number of rearing , no signifi-cance was performed(P >0.05).The levels of 5 -HT and 5 -HIAA in amygdala increased significantly (P <0.05). Though there was an increased trend of 5 -HIAA/5 -HT, no significance was detected by ANOVA(P >0.05).Con-clusion: The behavior and excitability of 5 -HT energetic nervous system in the amygdala of rats were changed by chron -ic restraint stress.JWGMDZT could effectively alleviate the impact of chronic restraint stress on rats .
