目的:观察槟榔十三味丸对慢性应激抑郁模型大鼠行为学和脑前额叶皮质、海马单胺类神经递质及其代谢产物的影响,探讨其抗抑郁作用机制。方法:将 Wistar 雄性大鼠,根据蔗糖水消耗量及体质量随机分为正常对照组、模型组、氟西汀组、槟榔十三味丸低、中、高剂量组,每组10只。除正常对照组外,其余大鼠均采用慢性轻度不可预见性应激结合孤养方法制备抑郁模型,造模同时灌胃给药,每日1次,连续给药21天。以敞箱实验、蔗糖水消耗实验进行行为学评价,并用高效液相-电化学法测定脑前额叶皮质、海马去甲肾上腺素(NE)、5-羟色胺(5-HT)、多巴胺(DA)及其代谢产物含量。结果:与正常对照组比较,模型组大鼠敞箱实验水平运动和垂直运动得分、蔗糖水消耗量显著降低(P <0.01或P <0.05);与模型组比较,槟榔十三味丸中、高剂量组和氟西汀组大鼠敞箱实验水平运动和垂直运动得分、蔗糖水消耗量均显著增加(P <0.05或 P <0.01);高效液相-电化学法测定结果显示,模型大鼠脑前额叶皮质 NE、5-HT、DA、DOPAC,海马5-HT、5-HIAA、DA 含量较正常对照组显著下降(P <0.05或 P <0.01),槟榔十三味丸可升高模型大鼠脑前额叶皮质 NE、5-HT、DA、DOPAC 和海马5-HT、5-HIAA、DA 含量,尤其中、高剂量组作用显著(P <0.05或 P <0.01)。结论:槟榔十三味丸能改善抑郁模型大鼠行为学异常,提高大鼠脑内神经递质 NE、5-HT、DA 及其代谢产物含量,这可能是槟榔十三味丸抗抑郁作用的重要机制之一。

목적:관찰빈랑십삼미환대만성응격억욱모형대서행위학화뇌전액협피질、해마단알류신경체질급기대사산물적영향,탐토기항억욱작용궤제。방법:장 Wistar 웅성대서,근거자당수소모량급체질량수궤분위정상대조조、모형조、불서정조、빈랑십삼미환저、중、고제량조,매조10지。제정상대조조외,기여대서균채용만성경도불가예견성응격결합고양방법제비억욱모형,조모동시관위급약,매일1차,련속급약21천。이창상실험、자당수소모실험진행행위학평개,병용고효액상-전화학법측정뇌전액협피질、해마거갑신상선소(NE)、5-간색알(5-HT)、다파알(DA)급기대사산물함량。결과:여정상대조조비교,모형조대서창상실험수평운동화수직운동득분、자당수소모량현저강저(P <0.01혹P <0.05);여모형조비교,빈랑십삼미환중、고제량조화불서정조대서창상실험수평운동화수직운동득분、자당수소모량균현저증가(P <0.05혹 P <0.01);고효액상-전화학법측정결과현시,모형대서뇌전액협피질 NE、5-HT、DA、DOPAC,해마5-HT、5-HIAA、DA 함량교정상대조조현저하강(P <0.05혹 P <0.01),빈랑십삼미환가승고모형대서뇌전액협피질 NE、5-HT、DA、DOPAC 화해마5-HT、5-HIAA、DA 함량,우기중、고제량조작용현저(P <0.05혹 P <0.01)。결론:빈랑십삼미환능개선억욱모형대서행위학이상,제고대서뇌내신경체질 NE、5-HT、DA 급기대사산물함량,저가능시빈랑십삼미환항억욱작용적중요궤제지일。
Objective:To study the effects on the behaviors and prefrontal cortex and hippocampus monoamine neurotrans -mitters and their metabolites in the chronic depressed rat models to probe the anti -depression mechanism of Mongolian pharmaceutical Betel Shisanwei ingredient pills .Methods:Sixty male Wistar rats were randomly divided into six groups according to the sugar consumption test and body weight (10 rats in each group):Normal control group,Model group, Fluoxetine group and High dose,medium dose and low dose groups of Betel Shisanwei ingredient pills .Except the normal control group,other groups were treated with the chronic unpredictable mild stress stimulation combined with lonely rai -sing for 21days.10mL/kg drugs were given to the rats,once daily,continuously for 21 days.After 21 days,behavior chan-ges of rats were detected through Open -field test,and sugar consumption test and body weight were also measured . HPLC -ECD techniques were used to analyze the levels of prefrontal cortex and hippocampus NE and 5 -HT and DA and their metabolites.Results:Compared with normal control group ,the scores of horizontal activity and vertical activity , sugar consumption decreased significantly in the model group (P <0.01 or P <0.05);Compared with model group,Flu-oxetine group and Betel Shisanwei ingredient pills group got significantly higher scores of horizontal activity ,vertical ac-tivity and sugar consumption (P <0.05 or P <0.01).HPLC -ECD techniques showed that the model rats prefrontal cortex NE,5 -HT,DA,DOPAC,hippocampus 5 -HT,5HIAA,DA content decreased more significantly than those in normal control group(P <0.05 or P <0.01).Betel Shisanwei ingredient pills could increase model rat 's prefrontal cortex NE,5 -HT,DA,DOPAC and hippocampus 5 -HT,5HIAA,DA content, especially the medium and high dose groups were significant(P <0.05 or P <0.01).Conclusion: Betel Shisanwei ingredient pills can improve abnormal behavior of depressive model rats, improve the neurotransmitter NE, 5 -HT, DA and its metabolic product content in rats brain, and it could be one of the important antidepressant effect mechanisms of Betel Shisanwei ingredient pills .
