目的:建立一种 TLC 生物自显影方法检测中药中单胺氧化酶抑制剂。方法:中药提取物以适当展开体系于硅胶板展开,然后以雾状均匀喷洒酶溶液,37℃预反应20min,之后以雾状喷洒显色剂与底物混合溶液。对于一个最优的底物选择,通过用 HPLC 来评价筛选一系列潜在底物化合物。结果:单胺氧化酶抑制剂在蓝色背景薄层板上会出现一个白色斑点,以磷酸异丙烟肼作为阳性药,检测限为0.01μg。中药路路通(Liquidambar formosana)乙醇提取物在硅胶板上展开显示出了单胺氧化酶抑制活性斑点。筛选出的最优底物为酪胺。结论:建立了一个新的 TLC 生物自显影方法,其可以快速筛选出中药中的单胺氧化酶抑制剂并为活性跟踪分离提供有力支持。

목적:건립일충 TLC 생물자현영방법검측중약중단알양화매억제제。방법:중약제취물이괄당전개체계우규효판전개,연후이무상균균분쇄매용액,37℃예반응20min,지후이무상분쇄현색제여저물혼합용액。대우일개최우적저물선택,통과용 HPLC 래평개사선일계렬잠재저물화합물。결과:단알양화매억제제재람색배경박층판상회출현일개백색반점,이린산이병연정작위양성약,검측한위0.01μg。중약로로통(Liquidambar formosana)을순제취물재규효판상전개현시출료단알양화매억제활성반점。사선출적최우저물위락알。결론:건립료일개신적 TLC 생물자현영방법,기가이쾌속사선출중약중적단알양화매억제제병위활성근종분리제공유력지지。
Ab stract:Objective: To develop a TLC bioautographic method to detect MAO inhibitors in traditional Chinese medicine . Method:After migration of the samples by proper solvent system , the TLC plate was sprayed with enzyme solution and in -cubated at 37℃ for 20 min, solutions of MTT and tyramine were mixed and sprayed onto the plate to give a blue back -ground colouration after 60 min.A series of compounds, as substrates, were purchased and their ability to be oxidized by MAO was evaluated by HPLC method.Results:Enzyme inhibitors (iproniazid or extracts) were observed as white spots on the TLC plates.Iproniazid inhibited MAO was down to 0.01 μg.Ethanol extracts of Liquidambar formosana af -ter migration on TLC showed inhibitive activity.Tyramine (2) was proved to be an appropriate substrate for TLC bioau -tographic assay.Conclusion:A TLC bioautographic method was developed for detecting the monoamine oxidase inhibitors in traditional Chinese medicine .
