Streptomyces sampsonii%Thiram-resistant%Mutation by UV irradiation
为选育抗药性桑氏链霉菌（ Streptomyces sampsonii），采用紫外线照射与药物培养基驯化结合法，筛选出MV-1、MV-2、MV-3、MV-4、MV-5等5株能在含有200 mg/L福美双的培养基上生长的抗性突变株。结果显示：出发原菌株KJ40对福美双最敏感，EC50为12．82 mg/L，致死浓度为40 mg/L。5株突变株中，MV-2和MV-5的EC50值最高，为129．28 mg/L，抗性水平最高，达10．09；MV-1最低，EC50为100．13 mg/L，抗性水平7．81。通过抑菌效果测定发现，MV-2和MV-4对紫丝核菌（ Rhizoctonia crocour m）抑菌力显著高于KJ40，抑菌率为80．39％和76．08％，分别比KJ40提高了10．59％和6．28％；MV-5无抑菌效果；MV-1和MV-3对病原菌的抑菌效果与K4J0无显著差异。拮抗活性高于亲本的MV-2和MV-4与福美双协同使用，抑菌效果高于菌株和福美双药液单独使用。
為選育抗藥性桑氏鏈黴菌（ Streptomyces sampsonii），採用紫外線照射與藥物培養基馴化結閤法，篩選齣MV-1、MV-2、MV-3、MV-4、MV-5等5株能在含有200 mg/L福美雙的培養基上生長的抗性突變株。結果顯示：齣髮原菌株KJ40對福美雙最敏感，EC50為12．82 mg/L，緻死濃度為40 mg/L。5株突變株中，MV-2和MV-5的EC50值最高，為129．28 mg/L，抗性水平最高，達10．09；MV-1最低，EC50為100．13 mg/L，抗性水平7．81。通過抑菌效果測定髮現，MV-2和MV-4對紫絲覈菌（ Rhizoctonia crocour m）抑菌力顯著高于KJ40，抑菌率為80．39％和76．08％，分彆比KJ40提高瞭10．59％和6．28％；MV-5無抑菌效果；MV-1和MV-3對病原菌的抑菌效果與K4J0無顯著差異。拮抗活性高于親本的MV-2和MV-4與福美雙協同使用，抑菌效果高于菌株和福美雙藥液單獨使用。
위선육항약성상씨련매균（ Streptomyces sampsonii），채용자외선조사여약물배양기순화결합법，사선출MV-1、MV-2、MV-3、MV-4、MV-5등5주능재함유200 mg/L복미쌍적배양기상생장적항성돌변주。결과현시：출발원균주KJ40대복미쌍최민감，EC50위12．82 mg/L，치사농도위40 mg/L。5주돌변주중，MV-2화MV-5적EC50치최고，위129．28 mg/L，항성수평최고，체10．09；MV-1최저，EC50위100．13 mg/L，항성수평7．81。통과억균효과측정발현，MV-2화MV-4대자사핵균（ Rhizoctonia crocour m）억균력현저고우KJ40，억균솔위80．39％화76．08％，분별비KJ40제고료10．59％화6．28％；MV-5무억균효과；MV-1화MV-3대병원균적억균효과여K4J0무현저차이。길항활성고우친본적MV-2화MV-4여복미쌍협동사용，억균효과고우균주화복미쌍약액단독사용。
We screened out five mutation strains resistant to thiram of MV-1, MV-2, MV-3, MV-4 and MV-5 by the combi-nation methods of UV irradiation and medical cultivation to select the thiram-resistant mutants of Streptomyces sampsonii.Among the tested strains of Streptomyces sampsonii, KJ40was the most sensitive to thiram , the EC50 was 12.82μg/mL, the lethal concentration was 40μg/mL.The resistant level of MV-2 and MV-5 were the highest between the thiram-resistant strains, and the EC50 and the resistant level were 129.28μg/mL and 10.09, respectively.The resistance level of MV-1 was the lowest, and the EC50 and the resistant level were 100.13μg/mL and 7.81, respectively.By determining inhibiting effect , inhibition activities of MV-2 and MV-4 were more significant than their parental strains , and the inhibition rate reached 80.39%and 76.08%, respectively.Compared with KJ40, the inhibition rates were improved by 10.59%and 6.28%, respectively. MV-5 had no antibacterial effect on Rhizoctoin a crocorum.Compared with the parental strains , there were no significant changes in inhibition activities of mv-1 and mv-3.By using the mixed of Thiram and Thiram-resistant mutants with the strongest inhibition activity , the inhibiting effect was more than that of thiram-resistant mutantsor and thiram alone .
