目的:探讨Nucleostemin基因(NS mRNA)表达检测对恶性葡萄胎的诊断意义。方法研究对象分为四组,A组为正常足月产孕妇,100例,采集羊水脱落细胞;B组为药物流产孕妇,100例,排除其他疾病,采集流产的胚胎绒毛组织; C组为良性葡萄胎患者,20例,采集葡萄胎组织和羊水脱落细胞;D组为恶性葡萄胎患者,19例,采集葡萄胎组织和羊水脱落细胞。采用Real-time PCR方法检测四组标本中Nucleostemin mRNA的表达水平。结果羊水脱落细胞和胚胎组织中Nucleostemin mRNA表达水平和阳性率以D组最高,C组次之。在羊水脱落细胞和胚胎组织中检测Nucleostemin mRNA,对于诊断恶性葡萄胎,敏感性、特异性和准确度均较好。结论 Nucleostemin基因高表达可能在葡萄胎的发生发展中起重要用,羊水脱落细胞和胚胎组织中Nucleostemin mRNA的检测有望成为恶性葡萄胎诊断的辅助指标。

목적:탐토Nucleostemin기인(NS mRNA)표체검측대악성포도태적진단의의。방법연구대상분위사조,A조위정상족월산잉부,100례,채집양수탈락세포;B조위약물유산잉부,100례,배제기타질병,채집유산적배태융모조직; C조위량성포도태환자,20례,채집포도태조직화양수탈락세포;D조위악성포도태환자,19례,채집포도태조직화양수탈락세포。채용Real-time PCR방법검측사조표본중Nucleostemin mRNA적표체수평。결과양수탈락세포화배태조직중Nucleostemin mRNA표체수평화양성솔이D조최고,C조차지。재양수탈락세포화배태조직중검측Nucleostemin mRNA,대우진단악성포도태,민감성、특이성화준학도균교호。결론 Nucleostemin기인고표체가능재포도태적발생발전중기중요용,양수탈락세포화배태조직중Nucleostemin mRNA적검측유망성위악성포도태진단적보조지표。
ObjectiveTo investigate the diagnosis significance of Nucleostemin for chorio-adenoma. MethodsThe patients were divided into four groups. Group A: 100 artifical abortion wemen. Group B:20 benign mole patients. Group C:19 chorio-adenoma patients. embryonic tissue and amniotic fluid cast-off cells were collected. Nucleostemin mRNA in the samples was detected by Real-time PCR.ResultsThe expression level and positive rate of Nucleostemin mRNA in amniotic fluid cast-off cells from Group C was the highest and group B was the second highest. The difference among the groups was significant.ConclusionDetection of the excessive expression of nucleostemin mRNA probably plays an important role in the diagnosis of benign mole and nucleostemin making it expression of a novel tumor marker in diagnosing chorio-adenoma.
