目的:观察三七总皂苷(panax notoginseng saponins,PNS)对兔骨髓间充质干细胞(bone marrow stromal cells, BMSCs)成骨分化及TGF-β1在基因水平表达变化的影响。方法:体外分离培养兔BMSCs,观察不同浓度PNS(50、100、200 mg/L)对兔BMSCs的影响,采用MTT法检测细胞增殖,茜素红染色观察钙结节形成,qRT-PCR检测TGF-β1表达水平。结果:各浓度PNS培养下的细胞增殖无明显差异(P>0.05);100、200 mg/L PNS组的钙结节数量和TGF-β1的表达水平均高于阴性组(P<0.05)。结论:PNS可促进兔BMSCs的成骨分化并刺激其分泌TGF-β1,但无明显促细胞增殖作用。

목적:관찰삼칠총조감(panax notoginseng saponins,PNS)대토골수간충질간세포(bone marrow stromal cells, BMSCs)성골분화급TGF-β1재기인수평표체변화적영향。방법:체외분리배양토BMSCs,관찰불동농도PNS(50、100、200 mg/L)대토BMSCs적영향,채용MTT법검측세포증식,천소홍염색관찰개결절형성,qRT-PCR검측TGF-β1표체수평。결과:각농도PNS배양하적세포증식무명현차이(P>0.05);100、200 mg/L PNS조적개결절수량화TGF-β1적표체수평균고우음성조(P<0.05)。결론:PNS가촉진토BMSCs적성골분화병자격기분비TGF-β1,단무명현촉세포증식작용。
Objective: To observe the osteoinduction effect of panax notoginseng saponins (PNS) on rabbit bone marrow mesenchymal stem cells (BMMSCs), and the expression level change of TGF-β1. Methods: Rabbit BMMSCs were isolated and cultured in vitro. To observe the influence of different concentration of PNS (50、100、200 mg/L) on rabbit BMMSCs, several methods were performed, including cell culture technique, MTT colorimetric assay, Dahl McGec Russell's Alizarin red staining for calcium and qRT-PCR explore TGF-β1 expression level. Results: There was no significant difference of proliferation capacity of BMMSCs between various concentration of PNS (P>0.05). The calcium nodule and TGF-β1 expression level in 100、200 mg/L PNS groups were significantly higher than negative group (P<0.05). Conclusion: PNS can promote the osteogenic differentiation and secretion of TGF-β1 of rabbit BMMSCs, but PNS is not effective enough for mediation of BMMSCs proliferation.
