目的:合成了一种新的稀土配合物[Nd(C20 H15 FN2 O4)2 Cl·H2 O]·7H2 O,并研究其抗肿瘤活性。方法:通过元素分析、摩尔电导、红外光谱、核磁共振氢谱和热重-差热分析,确定了该类配合物的化学组成。采用四氮唑蓝(MTT)比色法测定了配体 N1,N3-双邻甲苯甲酰基-氟尿嘧啶(DTFu)及配合物对 HepG2肝癌细胞株和 ZR-75-30人乳腺癌细胞株的抗肿瘤活性,流式细胞术检测肿瘤细胞周期。结果:DTFu 及其钕配合物对两种细胞株均有明显抑制增殖的能力,且钕配合物抗肿瘤活性明显强于配体。结论:配合物通过抑制细胞进入 S 期,影响肿瘤细胞内 DNA 的复制和合成,从而使细胞在 G1期堆积,最终抑制肿瘤细胞的增殖。

목적:합성료일충신적희토배합물[Nd(C20 H15 FN2 O4)2 Cl·H2 O]·7H2 O,병연구기항종류활성。방법:통과원소분석、마이전도、홍외광보、핵자공진경보화열중-차열분석,학정료해류배합물적화학조성。채용사담서람(MTT)비색법측정료배체 N1,N3-쌍린갑분갑선기-불뇨밀정(DTFu)급배합물대 HepG2간암세포주화 ZR-75-30인유선암세포주적항종류활성,류식세포술검측종류세포주기。결과:DTFu 급기녀배합물대량충세포주균유명현억제증식적능력,차녀배합물항종류활성명현강우배체。결론:배합물통과억제세포진입 S 기,영향종류세포내 DNA 적복제화합성,종이사세포재 G1기퇴적,최종억제종류세포적증식。
Obiective:To investigate the synthetic method and anti-tumor properties of the Nd(Ⅲ)complex[Nd(C20 H15 FN2 O4 )2 Cl·H2 O]·7H2 O. Methods:The structure was characterized by elemental analysis,molar conductance,IR spectra,1 H-NMR spectra and thermal analysis. MTT colorimetric assay was used to detect the anti-tumor properties of the ligand N1 ,N3-di-O-toluyl -fluorouracil and the complex on HepG2 human hepatocarcinoma cell line and ZR-75-30 hu-man breast cancer cell line. Cell cycle was detected by a flow cytometry. Results:It showed that the ligand and its complex had significant antiproliferative activities toward two tumor cell lines,and the anti-tumor property of the complex was markedly stronger than that of the ligand. Conclusion:Nd(Ⅲ)complex can inhibit cells entry into S period,affect the replication and synthesis of DNA in the tumor cells,so cells are accumulated in G1 period,and ultimately the complex in-hibits the proliferation of tumor cells.
