采用纸色谱、显色反应和紫外光谱对糯米藤中黄酮类化合物进行鉴别;运用Box-Behnken响应面法建立糯米藤总黄酮提取工艺条件的二次多项式数学模型,并对其抗氧化活性进行研究。结果表明:糯米藤中黄酮类化合物主要为黄酮醇类或黄酮类。利用响应面法优化出的工艺参数为:乙醇体积分数59.66%,提取温度77.2℃,提取时间1.4 h,液料比34 mL?g-1,在此条件下,糯米藤总黄酮实际得率达4.12%。所得模型拟合度高,试验误差小,可将此模型应用于糯米藤总黄酮提取的分析和预测。糯米藤总黄酮具有显著的DPPH自由基清除能力,其IC50为18.04μg?mL-1,是维生素C IC50的1.63倍;有较强的还原能力,约为维生素C的一半;具有一定的亚硝酸盐清除能力。

Flavonoid components from Gonostegia hirta were identified by paper chromatogram, color reaction and ultraviolet spectroscopy. A Box-Behnken design with a response surface methodology was used to study the flavonoid extraction conditions, and the antioxidantion activities of the flavonoids were also investigated. Results show that the main components of the flavonoids extracted are flavones and flavonols. The optimized extraction parameters are:ethanol volume fraction 59.66%, extraction temperature 77.2℃, extraction time 1.4 h and a liquid/solid ratio 34 mL?g-1. The flavonoids yield from Gonostegia hirta under the optimized condition is 4.12%. Moreover, the results show that the flavonoids from Gonostegia hirta have scavenging capacity against DPPH free radicals with an IC50 value of 18.04μg?mL-1, which is 1.63 times higher than that of Vitamin C. The reducing ability of studied flavonoids is approximately half of Vitamin C and the flavonoids also show nitrite cleaning ability.
