目的:观察咪喹莫特对正常包皮及疤痕疙瘩来源的原代培养成纤维细胞的增殖及凋亡的影响。方法分离培养正常包皮和疤痕疙瘩来源的成纤维细胞以及正常角质形成细胞,用5-溴脱氧尿嘧啶核苷(BrdU)掺入法检测不同浓度的咪喹莫特和经咪喹莫特4μg/ml刺激后的角质形成细胞培养上清液对两种组织来源的成纤维细胞增殖的影响,并以地塞米松作为药物对照;用流式细胞技术检测细胞凋亡。结果咪喹莫特和地塞米松均能抑制两种来源的成纤维细胞增殖( P<0.05),且咪喹莫特的抑制作用呈浓度依赖方式;咪喹莫特能显著提高角质形成细胞培养上清对疤痕来源成纤维细胞增殖的抑制作用( P<0.05);咪喹莫特、地塞米松以及经咪喹莫特刺激后的角质形成细胞上清液均无调节成纤维细胞凋亡的作用。结论咪喹莫特可直接抑制成纤维细胞增殖,还可通过刺激角质形成细胞间接抑制成纤维细胞增殖,但对成纤维细胞凋亡无明显影响。

Objective To investigate the effects of imiquimod on the proliferation and apopto-sis of primarily cultured fibroblasts that were derived from human keloid.Methods Human skin fi-broblasts were isolated from the foreskin and keloids , respectively , and cultured.The proliferation of the fibroblasts of the two origins was detected by MTT assay after treatment with various concentra -tions of imiquimod or with the supernatants of imiquimod-treated keratinocytes.The apoptosis of the fibroblasts was analyzed by flow cytometry.Dexamethasone was use in the study as a control drug.Results The proliferation of fibroblasts was significantly inhibited after treatment with imiquimod or dexamethasone ( P<0.05 ) .The inhibitory effect of imiquimod was in a dose-dependent manner.Imiquimod could significantly increase the inhibitory effect of the supernatants of keratinocytes on the proliferation of keloid-derived fibroblasts (P<0.05) .However, imiquimod, dexamethasone and the supernatants of imiquimod-treated keratimocytes failed to suppress the apoptosis of fibroblasts.Conclusion Imiquimod can inhibit the proliferation of fibroblasts directly or by stimulating kerati-nocytes, but it has no effect on the apoptosis of fibroblasts.
