目的:探讨卵巢切除后激素水平变化对肾脏组织水通道蛋白( AQP)表达和水代谢的影响。方法:60只大鼠随机分为2组。卵巢切除组大鼠切除卵巢后分别饲养0、2和4周( n=10);对照组大鼠只显露卵巢和输卵管,但不结扎切除,分别饲养0、2和4周( n=10)。分别收集2组尿、血液,取出肾脏,检测尿、血液渗透压,检测血浆雌、孕激素和血管加压素水平,免疫印迹和免疫组化检测肾脏组织AQP2、AQP1和AQP3的表达。结果:大鼠切除卵巢后2周,雌激素及孕激素水平均下降,血管加压素水平升高,体重增加,尿量减少,肾脏组织AQP2、AQP1和AQP3的表达上升(P均<0.05)。手术后4周,上述变化更加明显(P均<0.05)。结论:雌、孕激素水平下降可能通过血管加压素水平升高造成肾脏AQP高表达,进而导致尿量减少和体重增加。

목적:탐토란소절제후격소수평변화대신장조직수통도단백( AQP)표체화수대사적영향。방법:60지대서수궤분위2조。란소절제조대서절제란소후분별사양0、2화4주( n=10);대조조대서지현로란소화수란관,단불결찰절제,분별사양0、2화4주( n=10)。분별수집2조뇨、혈액,취출신장,검측뇨、혈액삼투압,검측혈장자、잉격소화혈관가압소수평,면역인적화면역조화검측신장조직AQP2、AQP1화AQP3적표체。결과:대서절제란소후2주,자격소급잉격소수평균하강,혈관가압소수평승고,체중증가,뇨량감소,신장조직AQP2、AQP1화AQP3적표체상승(P균<0.05)。수술후4주,상술변화경가명현(P균<0.05)。결론:자、잉격소수평하강가능통과혈관가압소수평승고조성신장AQP고표체,진이도치뇨량감소화체중증가。
Aim:To investigate the effects of decreased estrogen and progesterone on alteration of renal aquaporin ( AQP) and water retention after ovariectomy ( OVX) .Methods:A total of 60 rats were divided into 2 groups randomly .The expressions of AQP1,AQP2 and AQP3 in the kidney were examined at 0,2 and 4 weeks after operation in the two groups . Results:A persistent and significant increase in AQP 1,AQP2 and AQP3 expressions in the kidney at 2 and 4 weeks after OVX was observed ( P<0.05 ) .Plasma levels of estrogen and progesterone decreased ,whereas plasma AVP level was signif-icantly increased at 2 and 4 weeks after OVX(P<0.05).Urine output was significantly reduced while the weight of the rats was elevated at 2 and 4 weeks after OVX(P<0.05).Conclusion:Decreased estrogen and progesterone may contribute to the upregulation of renal AQP1,AQP2 and AQP3 through AVP pathway.The upregulation of AQP may play an important role in the decreased urine output and weight-loss after OVX.
