目的:观察加味三甲散对被动皮肤致敏型大鼠外周血单核细胞CD4+CD25+Treg mRNA表达的影响。方法:60只成年SD大鼠随机分为正常对照组,模型对照组,氯雷他定组和加味三甲散高、中、低剂量组,后5组均制作被动皮肤致敏模型。造模后氯雷他定组按10 mL/kg剂量灌胃给予氯雷他定水溶液,加味三甲散高、中、低剂量组分别按38.0、19.0、9.5 g/kg剂量灌胃给予加味三甲散提取浓缩液,正常对照组和模型对照组灌胃给予等体积纯净水。每天1次,连续3周。实验结束时,收集各组大鼠血液,采用实时荧光定量PCR技术检测CD4+CD25+Treg mRNA的表达。结果:6组大鼠外周血单核细胞 CD4+CD25+Treg mRNA 的表达水平差异有统计学意义( F =92.170,P<0.001);与正常对照组比较,模型对照组大鼠外周血单核细胞CD4+CD25+Treg mRNA的表达降低;与模型对照组比较,加味三甲散高、中、低剂量组大鼠外周血单核细胞CD4+CD25+Treg mRNA的表达升高( P均<0.05)。结论:加味三甲散可上调被动皮肤致敏型大鼠外周血单核细胞异常下降的CD4+CD25+Treg mRNA的表达水平,对慢性荨麻疹患者外周血单核细胞中CD4+CD25+Treg细胞功能的缺陷可能具有一定的干预作用。

목적:관찰가미삼갑산대피동피부치민형대서외주혈단핵세포CD4+CD25+Treg mRNA표체적영향。방법:60지성년SD대서수궤분위정상대조조,모형대조조,록뢰타정조화가미삼갑산고、중、저제량조,후5조균제작피동피부치민모형。조모후록뢰타정조안10 mL/kg제량관위급여록뢰타정수용액,가미삼갑산고、중、저제량조분별안38.0、19.0、9.5 g/kg제량관위급여가미삼갑산제취농축액,정상대조조화모형대조조관위급여등체적순정수。매천1차,련속3주。실험결속시,수집각조대서혈액,채용실시형광정량PCR기술검측CD4+CD25+Treg mRNA적표체。결과:6조대서외주혈단핵세포 CD4+CD25+Treg mRNA 적표체수평차이유통계학의의( F =92.170,P<0.001);여정상대조조비교,모형대조조대서외주혈단핵세포CD4+CD25+Treg mRNA적표체강저;여모형대조조비교,가미삼갑산고、중、저제량조대서외주혈단핵세포CD4+CD25+Treg mRNA적표체승고( P균<0.05)。결론:가미삼갑산가상조피동피부치민형대서외주혈단핵세포이상하강적CD4+CD25+Treg mRNA적표체수평,대만성담마진환자외주혈단핵세포중CD4+CD25+Treg세포공능적결함가능구유일정적간예작용。
Aim:To study the effect of modified Sanjiasan on the expression of CD 4 +CD25 +Treg mRNA in peripheral blood monocytes of rats with passive skin sensitization .Methods:A total of 60 rats were allocated into 6 groups:normal con-trol group,model control group,loratadine group,Sanjiasan high-,middle-and low-dose groups.The latter 5 groups were made passive skin sensitization model .The rats in the loratadine group were given 10 mL/kg loratadine by gavage ,the rats in Sanjiasan high-,middle-and low-dose groups were given 38.0,19.0,and 9.5 g/kg Sanjiasan,and the rats in normal control group and model control group were given normal saline ,once a day,for 3 weeks.Blood samples were collected to de-tect the expression of CD4 +CD25 +Treg mRNA by real time-PCR.Results:There were significant differences in the expres-sion of CD4 +CD25 +Treg mRNA among the 6 groups(F=92.170,P<0.001);compared with normal control,the expres-sion of CD4+CD25 +Treg mRNA in model control group decreased;compared with model control group ,the expression of CD4 +CD25 +Treg mRNA in Sanjiasan high-,middle-and low-dose groups elevated(P<0.05).Conclusion:Modified San-jiasan can upregulate the expression of CD 4 +CD25 +Treg mRNA that decreases in peripheral blood monocytes of rats with passive skin sensitization .Modified Sanjiasan may have certain intervention effect on the patients with chronic urticaria .
