采用荧光原位杂交技术(fluorescence in situ hybridization,FISH),以萝卜基因组特异序列pURsN作探针,分别与19份菜薹与萝卜附加系回交后代BC4A3d进行杂交,检测回交后代中外源萝卜染色体的存在情况。结果显示:只有1份材料BC4A3d-45没有杂交信号,其他18份材料均得到1个杂交信号,表明除了BC4A3d-45没有附加外源萝卜染色体之外,其余18份材料均含有1条外源萝卜染色体;该结果与分子标记鉴定结果一致。
採用熒光原位雜交技術(fluorescence in situ hybridization,FISH),以蘿蔔基因組特異序列pURsN作探針,分彆與19份菜薹與蘿蔔附加繫迴交後代BC4A3d進行雜交,檢測迴交後代中外源蘿蔔染色體的存在情況。結果顯示:隻有1份材料BC4A3d-45沒有雜交信號,其他18份材料均得到1箇雜交信號,錶明除瞭BC4A3d-45沒有附加外源蘿蔔染色體之外,其餘18份材料均含有1條外源蘿蔔染色體;該結果與分子標記鑒定結果一緻。
채용형광원위잡교기술(fluorescence in situ hybridization,FISH),이라복기인조특이서렬pURsN작탐침,분별여19빈채대여라복부가계회교후대BC4A3d진행잡교,검측회교후대중외원라복염색체적존재정황。결과현시:지유1빈재료BC4A3d-45몰유잡교신호,기타18빈재료균득도1개잡교신호,표명제료BC4A3d-45몰유부가외원라복염색체지외,기여18빈재료균함유1조외원라복염색체;해결과여분자표기감정결과일치。
Nineteen plants from the backcross progeny BC4A3d derived from backcrossing radish additional line with tai-tsai were analyzed by using fluorescence in situ hybridization(FISH)technology with radish genomic probe pURsN to detect the exogenous radish chromosome in the backcross individuals,so as to confirm the added radish chromosome in the backcross progenies.The results showed that except BC4A3d-45 had no additional exogenous radish chromosome,other 18 plants of the backcross progeny were added an alien radish chromosome. This result is consistent with that of the previous molecular marker identification.
