目的:研究载神经生长因子( NGF)纳米柔性脂质体的适宜制备的条件及稳定性。方法采用薄膜-超声法制备载NGF纳米柔性脂质体,ELISA法测定NGF含量,激光粒度分析仪检测粒径。结果制备载NGF纳米柔性脂质体的最适温度为30℃;pH值为7 R.0时,制备的纳米脂质体包封率最高;磷脂的适宜浓度为40 mg/ml;胆固醇与磷脂适宜比例为1∶4。室温保存30 d、4℃冰箱12个月,脂质体粒径无明显变化。结论本研究初步确定了载NGF纳米柔性脂质体的适宜制备条件,制备的脂质体具有较好稳定性。

목적:연구재신경생장인자( NGF)납미유성지질체적괄의제비적조건급은정성。방법채용박막-초성법제비재NGF납미유성지질체,ELISA법측정NGF함량,격광립도분석의검측립경。결과제비재NGF납미유성지질체적최괄온도위30℃;pH치위7 R.0시,제비적납미지질체포봉솔최고;린지적괄의농도위40 mg/ml;담고순여린지괄의비례위1∶4。실온보존30 d、4℃빙상12개월,지질체립경무명현변화。결론본연구초보학정료재NGF납미유성지질체적괄의제비조건,제비적지질체구유교호은정성。
Objective To study suitable preparation condition and stability of flexible nano-liposome ( FNL) loaded with nerve growth Factor ( NGF ) .Methods The FNL loaded with NGF was prepared by the method of filming and ultrasounding .The levels of NGF were detected by ELISA .The grain size of FNL was measured by laser grainsize analyzer . Results The optimal temperature of preparing FNL loaded with NGF was 35℃.The envelopment rate of prepared FNL peaked in pH 7.0.The suitable concentration of phosphatide was 40mg/ml.The best ratio of cholesterin:phosphatide was 1∶4.There was no obvious change in the grain size of FNL loaded with NGF after it was conserved in ambient temperature for 30 days and at 4℃in refrigerator for 12 months.Conclusion The study determines initially suitable preparation condition for FNL loaded with NGF,and prepared FNL has better stability .
