目的:研究补肾活血法中药(糖络通)对糖尿病大鼠早期视网膜病变的防治作用及其作用机制。方法:随机将33只正常雄性Wistar大鼠分为正常对照组(N)11只,糖尿病对照组(D)11只,糖络通治疗组(DT)11只,连续给药8周后观察大鼠体重、空腹血糖、FPG、糖化血红蛋白(HbA1c)及大鼠视网膜RAGE mRNA的表达水平。并通过光镜电镜观察大鼠视网膜组织病理形态学的变化。结果:实验研究表明,糖络通可降低糖尿病大鼠视网膜RAGE mRNA的表达水平,组间比较有差异性(P<0.01);ALT, AST,Cr,BUN等安全性指标未出现异常,组间比较无统计学差异(P>0.05)。结论:光镜、电镜观察表明糖络通对糖尿病大鼠的视网膜具有保护作用,能抑制神经节细胞及内外核层细胞的减少,减轻毛细血管扩张及视网膜组织的水肿。说明糖络通能够在一定程度上减缓DM大鼠视网膜的病理变化,对早期DR具有良好防治作用。为中医临床防治糖尿病微血管病变的研究提供理论依据。

목적:연구보신활혈법중약(당락통)대당뇨병대서조기시망막병변적방치작용급기작용궤제。방법:수궤장33지정상웅성Wistar대서분위정상대조조(N)11지,당뇨병대조조(D)11지,당락통치료조(DT)11지,련속급약8주후관찰대서체중、공복혈당、FPG、당화혈홍단백(HbA1c)급대서시망막RAGE mRNA적표체수평。병통과광경전경관찰대서시망막조직병리형태학적변화。결과:실험연구표명,당락통가강저당뇨병대서시망막RAGE mRNA적표체수평,조간비교유차이성(P<0.01);ALT, AST,Cr,BUN등안전성지표미출현이상,조간비교무통계학차이(P>0.05)。결론:광경、전경관찰표명당락통대당뇨병대서적시망막구유보호작용,능억제신경절세포급내외핵층세포적감소,감경모세혈관확장급시망막조직적수종。설명당락통능구재일정정도상감완DM대서시망막적병리변화,대조기DR구유량호방치작용。위중의림상방치당뇨병미혈관병변적연구제공이론의거。
Objective: To study the mechanism and preventive and therapeutic effects of Tangluo Tong on early retinopathy in diabetes rats. Methods:33 male rats were randomly divided into three groups, the control group (11 cases), the therapeutic group (11 cases) and the treatment group (11 cases), with 8 weeks’ medicine; the weight, the level of FPG, HbAlc and the expression of retinal RAGE mRNA in diabetes rats, and the histopathological changes were observed. Results: Tangluo Tong could reduce the expression level of retinal RAGE mRNA in diabetes rats with statistically significant difference (P<0.01);safe indicators such as ALT, AST, Cr, BUN were normal, there was no significant difference among groups (P>0.05). Tangluo Tong has protective effects for retina in diabetes rats, could inhibit the reduction of the ganglion cells and the inner and outer nuclear layer cells, and improve telangiectasias and retinal organization edema. Conclusion:The results suggest Tangluo Tong could improve pathological changes of retina in DM rats, prevent and treat early DR. All above provide theory basis for prevention and treatment study on diabetic microangiopathy in TCM.
