目的:研究壮筋续骨汤对大鼠股骨骨折后骨形态发生蛋白(BMP-7)和胰岛素样生长因子-1(IGF-1)血清水平的变化,探讨其促进骨折愈合的作用机制。方法成年健康雄性Wistar大鼠72只,用低速牙科钻在股骨中段切断股骨制备骨折模型,应用壮筋续骨汤水煎液干预治疗。X线观察骨折局部愈合情况,组织病理学观察骨痂结构,酶联免疫吸附试验检测血清BMP-7和IGF-1水平。结果大鼠骨折后7d骨折断端主要为纤维肉芽组织,14d为纤维-软骨性骨痂组织,21d骨性骨痂增多,模型组与治疗组未见显著差异,28d治疗组骨痂重塑质量明显优于模型组。大鼠术后7~21d,治疗组和模型组血清BMP-7和IGF-1水平无显著性差异,术后28d均显著降低(P<0.05),但治疗组明显高于模型组(P<0.05)。结论壮筋续骨汤可能通过降低内源性BMP-7和IGF-1降解,增强其活性,发挥促进骨折愈合的作用。

목적:연구장근속골탕대대서고골골절후골형태발생단백(BMP-7)화이도소양생장인자-1(IGF-1)혈청수평적변화,탐토기촉진골절유합적작용궤제。방법성년건강웅성Wistar대서72지,용저속아과찬재고골중단절단고골제비골절모형,응용장근속골탕수전액간예치료。X선관찰골절국부유합정황,조직병이학관찰골가결구,매련면역흡부시험검측혈청BMP-7화IGF-1수평。결과대서골절후7d골절단단주요위섬유육아조직,14d위섬유-연골성골가조직,21d골성골가증다,모형조여치료조미견현저차이,28d치료조골가중소질량명현우우모형조。대서술후7~21d,치료조화모형조혈청BMP-7화IGF-1수평무현저성차이,술후28d균현저강저(P<0.05),단치료조명현고우모형조(P<0.05)。결론장근속골탕가능통과강저내원성BMP-7화IGF-1강해,증강기활성,발휘촉진골절유합적작용。
Objective To investigate the effect of Zhuang Jin Xu Gu Decoction(ZJXG Decoction) on serum levels of bone morphogenetic protein-7(BMP-7)and insulin-like growth factor-1 (IGF-1)and to explore its promoting healing of femoral fracture in rats. Methods Femur fractures were generated in 72 male adult Wistar rats by cutting femur transversely at middle point. ZJXG Decoction was administered orally after surgery for 7-28 d. The healing process was analyzed by X-ray and hematoxtlin-eosin(HE)staining in rats. The serum levels of BMP-7 and IGF-1 were detected by enzyme linked immunobsorbent assay(ELISA). Results X-ray imaging indicated that the ifbrous callus tissue at the femoral fracture-end increased and the fracture line became fuzzy at 7-14 d following treatment with ZJXG Decoction. HE staining showed that the ifbrous-granular tissue at the fracture-end changed gradually to ifbrous, cartilaginous and osseous callus tissues. ELISA results showed that the serum levels of BMP-7 and IGF-1 increased signiifcantly 28 days following treatment with ZJXG Decoction.Conclusion It is concluded that ZJXG Decoction could enhance the fracture healing by reducing the decomposition of BMP-7 and IGF-1 and enhancing their activities.
