ANXA2基因属于膜联蛋白家族一员,N-末端区域包含调控PKC通路中酪氨酸和色氨酸两种蛋白激酶磷酸化的位点,曾有研究发现ANXA2基因在高产蛋鸡卵巢中出现了显著性高表达。因此,本实验利用荧光定量PCR方法,研究了ANXA2基因在鸡卵巢和各等级卵泡中的表达规律,发现此基因在开产鸡卵巢中表达升高,并随卵泡发育表达上调,排卵后表达量出现显著性下降。通过分离培养鸡卵泡的膜细胞,添加促卵泡生长素(FSH)和促黄体素(LH)处理,提取细胞RNA和利用荧光定量PCR方法,发现此基因的表达升高与FSH的调控有关,却与LH无关。此作用的具体分子机制,还待于今后进一步的研究。

ANXA2 is a family member of the membrane-bound proteins. The N-terminal region contains the tyrosine and threonine phosphorylation sites for protein kinase activation in PKC pathway regulations. Previous studies showed that ANXA2 gene is overexpressed in hens with high ovulation efficiency. This paper has studied the expression patters of ANXA2 gene during the chicken ovary and follicles development by the real-time PCR. The results showed that the expression of ANXA2 increased with the chicken ovary maturation. In hierarchy follicles, ANXA2 mRNA increased significantly as follicular development proceeds in pre-ovulatory follicles, reached the peak expression at the time of ovulation then decreased significantly after ovulation. Then, the theca cells of pre-ovulatory follicles were isolated, and treated with different concentrations of FSH or LH. We found that the theca cell ANXA2 expression was stimulated by FSH but not LH. The molecular mechanism of the ANXA2 regulation in chicken ovary should be studied further.
