目的:用戊四氮(PTZ)诱导建立的慢性癫痫(EP)幼鼠模型,研究神经生长因子(NGF)处理后幼鼠海马细胞色素C(Cyt C)的表达,探讨 NGF在 EP发病中的作用。方法50只SD幼鼠随机分为阴性对照组(A组)、阳性对照组(B组)、NGF低剂量组(C组)、NGF中剂量组(D组)、NGF高剂量组(E组)。NGF用药后3 d ,取海马组织用于免疫组织化学及实时荧光定量PCR方法检测Cyt C的表达。结果与A组比较,B组Cyt C阳性细胞数及mRNA 表达量明显增多(P<0.01)。与B组比较,C~ E组Cyt C阳性细胞数及 mRNA 表达量降低(P<0.05)。结论 NGF可减少海马Cyt C的表达,促进EP幼鼠体质量增长。

목적:용무사담(PTZ)유도건립적만성전간(EP)유서모형,연구신경생장인자(NGF)처리후유서해마세포색소C(Cyt C)적표체,탐토 NGF재 EP발병중적작용。방법50지SD유서수궤분위음성대조조(A조)、양성대조조(B조)、NGF저제량조(C조)、NGF중제량조(D조)、NGF고제량조(E조)。NGF용약후3 d ,취해마조직용우면역조직화학급실시형광정량PCR방법검측Cyt C적표체。결과여A조비교,B조Cyt C양성세포수급mRNA 표체량명현증다(P<0.01)。여B조비교,C~ E조Cyt C양성세포수급 mRNA 표체량강저(P<0.05)。결론 NGF가감소해마Cyt C적표체,촉진EP유서체질량증장。
Objective To explore the effects of nerve growth factor (NGF) on the expression of cytochrome c(Cyt C) in hippocampus of pentylenetetrazol (PTZ)-induced epileptic rats . Methods Fifty SD rats were randomly divided into 5 groups :negative control group (group A ) ,positive control group (group B) ,NGF low-dose group (group C) ,NGF medium-dose group (group D) and NGF high-dose group (group E) . After 3 days of NGF treatment ,the number of Cyt C positive cells and the expression of Cyt C mRNA in hippocampus were detected by immunohistochemisty and real-time PCR . Results The number of Cyt C positive cells and the expression of Cyt C mRNA in hippocampus of group B were significantly higher than group A (P<0 .01) . The number of Cyt C positive cells and the expression of Cyt C mRNA of group C~E were lower than group B (P<0 .05) . Conclusion NGF may play an impor-tant role in decreasing the expression of Cyt C and increasing the weight of epileptic rats .
