目的:通过检测人口腔鳞状细胞癌(OSCC)中血小板源性生长因子-A(PDGF-A)及血小板源性生长因子-A 受体-α(PDGFR-α)和血小板-内皮细胞黏附分子(CD31)蛋白标记的微血管密度(MVD),探讨 PDGF-A 及 PDGFR-α在 OSCC 中的作用及与血管生成的关系,并进一步阐述其临床指导意义。方法收集贵阳医学院附属医院2011年10月至2013年10月手术切除的 OSCC 标本42例(研究组)以及正常口腔黏膜组织11例(对照组),采用免疫组化(SABC)法分别检测两组的 PDGF-A、PDGFR-α和 CD31表达并计数 MVD。结果(1)PDGF-A 在研究组和对照组中的表达率分别为73.81%、9.09%,而 PDGFR-α则分别为78.57%、9.09%,差异均有统计学意义(均 P<0.05)。(2)在 OSCC 中,PDGF-A 的表达与肿瘤分化程度及是否发生淋巴结转移相关(P<0.05),而 PDGFR-α仅与肿瘤分化程度相关(P<0.05)。(3)在研究组中, PDGF-A、PDGFR-α的阳性表达者 MVD 值均高于阴性者,差异均有统计学意义(均 P<0.05)。结论 OSCC 中 PDGF-A、PDGFR-α可能通过血管生成参与了肿瘤的生长和转移,但其在口腔鳞癌中的作用机制还有待进一步研究。

목적:통과검측인구강린상세포암(OSCC)중혈소판원성생장인자-A(PDGF-A)급혈소판원성생장인자-A 수체-α(PDGFR-α)화혈소판-내피세포점부분자(CD31)단백표기적미혈관밀도(MVD),탐토 PDGF-A 급 PDGFR-α재 OSCC 중적작용급여혈관생성적관계,병진일보천술기림상지도의의。방법수집귀양의학원부속의원2011년10월지2013년10월수술절제적 OSCC 표본42례(연구조)이급정상구강점막조직11례(대조조),채용면역조화(SABC)법분별검측량조적 PDGF-A、PDGFR-α화 CD31표체병계수 MVD。결과(1)PDGF-A 재연구조화대조조중적표체솔분별위73.81%、9.09%,이 PDGFR-α칙분별위78.57%、9.09%,차이균유통계학의의(균 P<0.05)。(2)재 OSCC 중,PDGF-A 적표체여종류분화정도급시부발생림파결전이상관(P<0.05),이 PDGFR-α부여종류분화정도상관(P<0.05)。(3)재연구조중, PDGF-A、PDGFR-α적양성표체자 MVD 치균고우음성자,차이균유통계학의의(균 P<0.05)。결론 OSCC 중 PDGF-A、PDGFR-α가능통과혈관생성삼여료종류적생장화전이,단기재구강린암중적작용궤제환유대진일보연구。
Objective By detecting human oral squamous cell carcinoma (OSCC) in platelet-derived growth factor -A (PDGF-A) and platelet-derived growth factor receptor -α (PDGFR-α) and platelet - endothelial cell adhesion molecule (CD31) protein marker of microvessel density(MVD), to explore the role and clinical significance of PDGF-A and PDGFR-α in OSCC and its relationship with angiogenesis. Methods From October 2011 to October 2013,42 cases of resected specimens of OSCC(the study group) and 11 cases of normal oral mucosa(the control group) were collected in the Affiliated Hospital of Guiyang Medical College,immunohistochemistry (SABC) were used to detect the expression of PDGF-A,PDGFR-αand CD31 expression and take count of the number of MVD in the two groups. Results 1.Positive expression of PDGF-A in the control group and study group were 73.81% and 9.09% respectively,positive expression of PDGFR-αwere 78.57% and 9.09%,there was significant difference(all P<0.05).The MVD in the study group were significantly higher than that in the control group(P<0.05).2.The expression of PDGF-A was correlated with TNM grade and lymph node metastasis in OSCC(P<0.05),and the expression of PDGFR-αwas only correlated with TNM grade in OSCC(P<0.05).3.The MVD in the positive expression of PDGF-A and PDGFR-α were higher than that in negative in OSCC,there was significant difference(all P<0.05). Conclusion PDGF-A and PDGFR-α may take part in the growth,progression and metastasis through angiogenesis in OSCC,but their specific mechanism need further research.
