Heteropappus altaicus (Willd.) Novopokr%ultrasonic extraction%polysaccharide%content determination
目的:采用超声法提取阿尔泰狗娃花中的多糖,并测定其含量。方法:采用紫外-可见分光光度法,以葡萄糖为对照品,苯酚-浓硫酸显色法测定阿尔泰狗娃花中多糖的含量。结果:总多糖在2.535-15.21μg/mL范围内呈良好线性关系,回归方程Y=0.0501X-0.0015(r=0.9998,n=6),阿尔泰狗娃花多糖含量为5.645%, RSD为0.65%。经方法学考察显示,稳定性、精密度、重现性均较好,平均回收率为96.86%,RSD为1.45%。结论:紫外分光光度法测定阿尔泰狗娃花中的多糖,操作简便、灵敏度高、重现性好,稳定性高,可作为阿尔泰狗娃花中多糖含量测定方法。

목적:채용초성법제취아이태구왜화중적다당,병측정기함량。방법:채용자외-가견분광광도법,이포도당위대조품,분분-농류산현색법측정아이태구왜화중다당적함량。결과:총다당재2.535-15.21μg/mL범위내정량호선성관계,회귀방정Y=0.0501X-0.0015(r=0.9998,n=6),아이태구왜화다당함량위5.645%, RSD위0.65%。경방법학고찰현시,은정성、정밀도、중현성균교호,평균회수솔위96.86%,RSD위1.45%。결론:자외분광광도법측정아이태구왜화중적다당,조작간편、령민도고、중현성호,은정성고,가작위아이태구왜화중다당함량측정방법。
Objective:To extract the polysaccharide of Heteropappus altaicus( Willd. ) novopokr by ultrasonic extraction, and determine the polysaccharide content. Methods:Based on the standard sample glucose,the polysaccharide content of Heteropappus altaicus ( Willd. ) novopokr polysaccharide was determined by phenol-sulfuric acid coloration and Ultraviolet and Visible( UV) spectrophotometry. Results:The results showed that the absorbance has a linear relationship with the concentration in the range of 2. 535~15. 21μg/mL,and the regression equation is Y=0. 0501X-0. 0015(r=0. 9998,n=6). The polysaccharide consent determined is 5. 645% with 0. 65% of RSD. The methodological evaluation demonstrated that this method has good stability, precision and reproducibility. Also, the average recovery rate is 96 . 86%and RSD is 0 . 65%. Conclusion:The UV-spectrophotometry can be employed to determine the polysaccharide content of Heteropappus altaicus( Willd. ) novopokr,because the method is convenient in operation and has high sensitivity,good repeatability and stability.
