Rubus amabilis%polysaccharide%arabinogalactan%immunobiological activity
目的:研究植物秀丽莓(Rubus amabilis)茎中分离得到的多糖RAP-B-1的理化性质以及免疫活性。方法秀丽莓茎经过水煎、醇沉、透析获得粗多糖(crude polysaccharide,RAP)。RAP再经DEAE-纤维素和Sephadex G-100柱色谱分离得到相对分子质量均一的多糖RAP-B-1。采用完全酸水解、高碘酸氧化、Smith降解及甲基化反应,以及毛细管电泳(capillary electrophoresis,CE)、红外(infrared radiation,IR)、磁共振(nuclear magnetic resonance,NMR)和气相色谱-质谱联用(gas chromatography-mass spectrometry,GC-MS)等方法研究其理化性质。采用小鼠脾淋巴细胞增殖和腹腔巨噬细胞吞噬实验评价RAP-B-1的免疫活性。结果多糖RAP-B-1的峰位相对分子质量为4.80×104,比旋光度为[α]20D +68.3(c=1,H2O),其单糖组成为木糖:阿拉伯糖:葡萄糖:鼠李糖:半乳糖:甘露糖:葡萄糖醛酸:半乳糖醛酸=1.0:6.9:0.8:1.1:6.9:0.3:0.5:3.3,推测RAP-B-1主要为酸性阿拉伯半乳聚糖,其中阿拉伯糖的连接方式:→1) Ara(2,3→、→1) Ara(5→和→1) Ara;半乳糖的连接方式:→1)Gal(4→、→1)Gal(6→和→1)Gal。生物活性评价结果表明RAP-B-1在1~50μg/ml范围内对小鼠脾脏T细胞有一定促增殖作用(P <0.05),同时增强小鼠腹腔巨噬细胞的吞噬能力(P <0.01)。结论RAP-B-1为一种阿拉伯半乳聚糖,具有一定免疫促进作用。

목적:연구식물수려매(Rubus amabilis)경중분리득도적다당RAP-B-1적이화성질이급면역활성。방법수려매경경과수전、순침、투석획득조다당(crude polysaccharide,RAP)。RAP재경DEAE-섬유소화Sephadex G-100주색보분리득도상대분자질량균일적다당RAP-B-1。채용완전산수해、고전산양화、Smith강해급갑기화반응,이급모세관전영(capillary electrophoresis,CE)、홍외(infrared radiation,IR)、자공진(nuclear magnetic resonance,NMR)화기상색보-질보련용(gas chromatography-mass spectrometry,GC-MS)등방법연구기이화성질。채용소서비림파세포증식화복강거서세포탄서실험평개RAP-B-1적면역활성。결과다당RAP-B-1적봉위상대분자질량위4.80×104,비선광도위[α]20D +68.3(c=1,H2O),기단당조성위목당:아랍백당:포도당:서리당:반유당:감로당:포도당철산:반유당철산=1.0:6.9:0.8:1.1:6.9:0.3:0.5:3.3,추측RAP-B-1주요위산성아랍백반유취당,기중아랍백당적련접방식:→1) Ara(2,3→、→1) Ara(5→화→1) Ara;반유당적련접방식:→1)Gal(4→、→1)Gal(6→화→1)Gal。생물활성평개결과표명RAP-B-1재1~50μg/ml범위내대소서비장T세포유일정촉증식작용(P <0.05),동시증강소서복강거서세포적탄서능력(P <0.01)。결론RAP-B-1위일충아랍백반유취당,구유일정면역촉진작용。
Objective To investigate the physicochemica l properties and immunobiological activity of a polysaccharide (RAP-B-1) from stems of Rubus amabilis. Methods The crude polysaccharide (RAP) was obtained successively by boiling, ethanol precipitating and dialyzing. RAP was isolated with DEAE-cellulose and Sephadex G-100 to obtain a polysaccharide RAP-B-1. The physicochemical properties of RAP-B-1 were studied by hydrolysis, periodate oxidation, Smith degradation and methylation, CE, IR, NMR and GC-MS. The immunobiological activities were estimated by the proliferative activity of spleen lymphocytes and phagocytic activity of peritoneal macrophages in mice. Results The molecular weight of RAP-B-1 was 4.80×104 with specific optical rotation value [α] 20D+68.3 (c=1,H2O), and was composed of eight monosaccharides. The molar ratios were as Xyl: Ara: Glc: Rha:Gal: Man: GlcA: GalA = 1.0:6.9:0.8:1.1:6.9:0.3:0.5:3.3. RAP-B-1 was an arabinogalactan. The linkages of arabinose were →1) Ara (2,3→,→1) Ara(5→and→1) Ara, and the linkages of galactose were→1) Gal(4→,→1) Gal(6→and→1) Gal. RAP-B-1 could improve the proliferative activity of spleen T cells(P<0.05) and booste phagocytic activity of peritoneal macrophages at 50μg/ml concentration(P<0.01). Conclusion RAP-B-1 is an arabinogalactan and has immunobiological activity.
