应用PC R技术对A型产气荚膜梭菌青海分离株的α毒素基因进行扩增，并进行测序，利用生物软件对α毒素基因序列进行分析，对蛋白结构、B细胞抗原表位进行预测。结果表明，A型产气荚膜梭菌青海分离株的α毒素基因长度为1110 bp ，编码370个氨基酸。与参考菌株T0、S16、NRRLB-23744、KZ211和CER89L1216的α毒素基因核苷酸序列同源性依次为99．9％、99．9％、99．9％、99．9％和99．7％，氨基酸序列同源性均为100％。综合α毒素蛋白亲水性、表面可能性、抗原指数及二级结构预测结果显示在26-29、71-78、111-114、182-185、189-192、264-268、272-275、282-285、294-297、362-365位氨基酸残基存在可能的B细胞抗原表位，结合三级结构预测结果显示，α毒素294位-297位氨基酸残基形成表位并结合抗体的可能性较高。研究结果可为A型产气荚膜梭菌表位疫苗的设计与研制提供基础。

α toxin gene of Clostridium per f ringens type A was amplifed by PCR and sequenced .The gene sequence ,protein structure and epitope of α toxin were analyzed and predicted .The result showed that αtoxin gene length was 1 110 bp ,encoding 370 amino acids .Nucleotide sequence homology of αtoxin gene of reference strains T0 ,S16 ,NRRL B-23744 ,KZ211 ,CER 89L1216 were 99 .9% ,99 .9% ,99 .9% , 99 .9% and 99 .7% ,amino acid sequence homology were 100% .Predictions of hydrophilicity ,surface possi-bilities ,antigen index and secondary structure showed in 26-29 ,71-78 ,111-114 ,182-185 ,189-192 ,264-268 , 272-275 ,282-285 ,294-297 ,362-365 of αtoxin existed B cell antigen epitopes ,binding tertiary structure pre-diction showed there had the higher possibility of epitopes in 294-297 amino acid residues of α toxin .The research results can provide a basis for design and development epitode vaccine of Clostridium per f ringens type A .
