为建立一种检测肉制品中犬源性成分的PCR检测方法,用于生、熟犬肉的鉴定,采用PCR方法特异性扩增生、熟犬肉基因组DNA ,并采用 PCR方法检测46份保定市的犬肉制品。所检犬源性成分在213 bp处出现预期的DNA目的片段,扩增片段经限制性内切酶 H phⅠ酶切分析确认,所得179 bp和34 bp片段与预期目标一致。应用该引物均可扩增出犬的目的D N A片段,而对马、山羊、绵羊、驴、兔和猪等15种动物肉的DN A扩增则呈阴性。其检测灵敏度达到4.56 pg DN A。利用本法对46份犬肉制品进行检测,检出率为100%。试验表明建立的方法快速简便,且具有较高的特异性和敏感性,用于生、熟犬肉的鉴定是可行的。

To establish a method for detecting canine ingredient in raw and cooked canine meat by polymer-ase chain reaction(PCR) ,the genomic DNA extracted from canine samples was amplified by PCR for de-tection of raw and cooked canine meat ,and the 46 meat products were detected using PCR .A pair of spe-cial primer were chosen to amplified 213 bp fragment .A H phⅠrestriction endonuclease test was simulta-neously made for verifying the specificity of the PCR .The results showed that a specific amplification frag-ment of the expected size was detected and demonstrated that it was positive for canine meat ,but negative for equine ,goat ,ovine ,donkey ,rabbit and swine meat etc .The detection limit was 4 .56 fg DNA .The 46 meat products of dogs were detected using PCR ,and the positive rate was 100% .Experiments showed that the method had higher ,sensitivity and specificity for raw and cooked canine meat identification .
