目的:观察脓毒症大鼠血脑屏障通透性及脑组织胞质附着蛋白( ZO-1)的变化。方法50只健康SD大鼠随机分为假手术组( n=10)及脓毒症模型组( n=40),脓毒症模型组又分为脓毒症制模后6、12、24、48 h 4个亚组,每组10只,采用盲肠结扎穿孔( CLP)法制作脓毒症模型。于相应时间点处死取材,伊文思蓝( EB)外渗分光光度分析法检测血脑屏障通透性;RT-PCR法测定脑组织ZO-1 mRNA表达、Western-blot免疫印迹方法检测ZO-1表达。结果脓毒症组各时间点大鼠脑内EB含量均明显高于假手术组( P<0.01);脓毒症各时间点组大鼠脑内ZO-1 mRNA、ZO-1的表达均明显下降,与假手术组比较差异有统计学意义(P<0.01),且随着脓毒症时间延长下降越明显。结论脓毒症大鼠脑内ZO-1 mRNA、ZO-1的表达均显著下降,可能是脓毒症血脑屏障受损的机制之一。
目的:觀察膿毒癥大鼠血腦屏障通透性及腦組織胞質附著蛋白( ZO-1)的變化。方法50隻健康SD大鼠隨機分為假手術組( n=10)及膿毒癥模型組( n=40),膿毒癥模型組又分為膿毒癥製模後6、12、24、48 h 4箇亞組,每組10隻,採用盲腸結扎穿孔( CLP)法製作膿毒癥模型。于相應時間點處死取材,伊文思藍( EB)外滲分光光度分析法檢測血腦屏障通透性;RT-PCR法測定腦組織ZO-1 mRNA錶達、Western-blot免疫印跡方法檢測ZO-1錶達。結果膿毒癥組各時間點大鼠腦內EB含量均明顯高于假手術組( P<0.01);膿毒癥各時間點組大鼠腦內ZO-1 mRNA、ZO-1的錶達均明顯下降,與假手術組比較差異有統計學意義(P<0.01),且隨著膿毒癥時間延長下降越明顯。結論膿毒癥大鼠腦內ZO-1 mRNA、ZO-1的錶達均顯著下降,可能是膿毒癥血腦屏障受損的機製之一。
목적:관찰농독증대서혈뇌병장통투성급뇌조직포질부착단백( ZO-1)적변화。방법50지건강SD대서수궤분위가수술조( n=10)급농독증모형조( n=40),농독증모형조우분위농독증제모후6、12、24、48 h 4개아조,매조10지,채용맹장결찰천공( CLP)법제작농독증모형。우상응시간점처사취재,이문사람( EB)외삼분광광도분석법검측혈뇌병장통투성;RT-PCR법측정뇌조직ZO-1 mRNA표체、Western-blot면역인적방법검측ZO-1표체。결과농독증조각시간점대서뇌내EB함량균명현고우가수술조( P<0.01);농독증각시간점조대서뇌내ZO-1 mRNA、ZO-1적표체균명현하강,여가수술조비교차이유통계학의의(P<0.01),차수착농독증시간연장하강월명현。결론농독증대서뇌내ZO-1 mRNA、ZO-1적표체균현저하강,가능시농독증혈뇌병장수손적궤제지일。
Obj ective To observe the changes on blood -brain barrier permeability and brain expression of ZO -1 in sepsis rats.Methods Fifty SD rats were divided into sham operation group (n=10) and model group (n =40).The model rats were further divided into 6, 12, 24, 48 h subgroups (n=10).The sepsis rat model was developed by CLP .The blood-brain barrier permeability was detected by spectrophotometric analysis of EB .The expression of ZO -1 mRNA and protein was detected by RT-PCR and Western -blot, respectively.Results The EB contents of each time point groups were higher than sham operation group (P<0.01).The expression of ZO-1 in each time point groups were lower than sham operation group (P <0.01).And the level of ZO -1 expression was continuously decreased during the development of sepsis .Conclusion The expression levels of ZO -1 mRNA and protein in sepsis rats are decreased significantly , and this may be one of the mechanisms of blood-brain barrier damage of sepsis .
