目的:通过观察虎杖干预后肺纤维化大鼠肺组织中超氧化物歧化酶(SOD)活性及谷胱甘肽(GSH)、丙二醛(MDA)含量的影响,探讨虎杖防治肺纤维化的作用机制。方法健康雄性SD大鼠120只,随机分为5组:模型组30只,气管内一次性注入博莱霉素50.5mg/100g制作肺纤维化大鼠模型,当日予0.9%氯化钠溶液5ml/100g灌胃;预防组30只,于造模前2d开始予虎杖5ml/100g灌胃;肺泡炎组20只,于造模后第7天开始虎杖灌胃;肺纤维化组10只,于造模后第28天开始虎杖灌胃;对照组30只,于造模当日开始0.9%氯化钠溶液5ml/100g灌胃。各组大鼠灌胃均1次/d。5组大鼠分别于第3、7、14、28、42或56天分阶段处死5只,取肺组织制成匀浆,测定SOD活性及GSH、MDA的含量。结果模型组第3、7、14天肺组织中SOD活性及GSH含量低于对照组(均P<0.05),预防组上述指标较模型组升高(P<0.05或0.01),肺泡炎组、肺纤维化组与模型组的差异均无统计学意义(均P>0.05)。模型组第7、14天肺组织中MDA含量明显高于对照组和预防组(均P<0.05)。结论虎杖能通过增强肺组织局部抗氧化能力,减轻博莱霉素诱导的肺泡炎和肺纤维化程度,可能是治疗肺纤维化的有效药物。

목적:통과관찰호장간예후폐섬유화대서폐조직중초양화물기화매(SOD)활성급곡광감태(GSH)、병이철(MDA)함량적영향,탐토호장방치폐섬유화적작용궤제。방법건강웅성SD대서120지,수궤분위5조:모형조30지,기관내일차성주입박래매소50.5mg/100g제작폐섬유화대서모형,당일여0.9%록화납용액5ml/100g관위;예방조30지,우조모전2d개시여호장5ml/100g관위;폐포염조20지,우조모후제7천개시호장관위;폐섬유화조10지,우조모후제28천개시호장관위;대조조30지,우조모당일개시0.9%록화납용액5ml/100g관위。각조대서관위균1차/d。5조대서분별우제3、7、14、28、42혹56천분계단처사5지,취폐조직제성균장,측정SOD활성급GSH、MDA적함량。결과모형조제3、7、14천폐조직중SOD활성급GSH함량저우대조조(균P<0.05),예방조상술지표교모형조승고(P<0.05혹0.01),폐포염조、폐섬유화조여모형조적차이균무통계학의의(균P>0.05)。모형조제7、14천폐조직중MDA함량명현고우대조조화예방조(균P<0.05)。결론호장능통과증강폐조직국부항양화능력,감경박래매소유도적폐포염화폐섬유화정도,가능시치료폐섬유화적유효약물。
Objective To evaluate the effects Rhizoma Polygoni Cuspidati on SOD,GSH and MDA contents in lung tissue of rats with bleomycetin- induced pulmonary fibrosis. Methods One hundred and twenty healthy male SD rats were randomly divided in to 5 groups. Pulmonary fibrosis model was induced by intratracheal infusion of 50.5mg/100g bleomycetin. Rats in model group (n=30) were instil ed with saline 5ml/d by gavage after bleomycetin was infused;rats in preventive group(n=30)were treated with liquid extract of Rhizoma Polygoni Cuspidati 2d before bleomycetin infused;rats in alveolitis group (n=20)and pul-monary fibrosis group (n=10) were treated with liquid extract of Rhizoma Polygoni Cuspidati 7d after bleomycetin infused;rats in control group (n=30) were instil ed with saline 5ml/d. Five rats in each group were sacrificed on d3,7,14,28,42 and 56, and lung samples were col ected. The contents of SOD,GSH and MDA in lung homogenate were assayed. Results The levels of SOD and GSH on d 3,7 and 14 were lower in model group than those in control group(P<0.05) and preventive group(P<0.05);the levels of MDA on d7 and 14 were higher in model group than those in control group and preventive group (P<0.05). Conclusion Liquid extract of Rhizoma Polygoni Cuspidati has anti- oxidant effects, which is associated with the al eviation of alveolitis pulmonary fi-brosis induced by bleomycetin in rats.
