目的:研究吲哚美辛对受损颈动脉再内皮化的作用及机制。方法在SD大鼠体内通过球囊法制备颈动脉损伤模型,腹腔注射吲哚美辛,观察其对血管内皮修复的影响;密度梯度离心法获得大鼠单核细胞,应用EGM-2培养基体外诱导培养获得内皮祖细胞( EPCs)。采用不同浓度的吲哚美辛处理晚期EPCs24 h后,MTT法检测细胞的增殖活性,细胞差异贴壁法检测 EPCs的粘附能力,改良的 Boyden小室检测 EPCs的迁移能力,Matrigel基质胶检测EPCs的成血管能力。结果吲哚美辛可明显促进损伤血管内皮的修复,其机制可能与吲哚美辛增强 EPCs的增殖、粘附、迁移及成血管能力有关。结论吲哚美辛促进了损伤血管内皮的再内皮化进程。

목적:연구신타미신대수손경동맥재내피화적작용급궤제。방법재SD대서체내통과구낭법제비경동맥손상모형,복강주사신타미신,관찰기대혈관내피수복적영향;밀도제도리심법획득대서단핵세포,응용EGM-2배양기체외유도배양획득내피조세포( EPCs)。채용불동농도적신타미신처리만기EPCs24 h후,MTT법검측세포적증식활성,세포차이첩벽법검측 EPCs적점부능력,개량적 Boyden소실검측 EPCs적천이능력,Matrigel기질효검측EPCs적성혈관능력。결과신타미신가명현촉진손상혈관내피적수복,기궤제가능여신타미신증강 EPCs적증식、점부、천이급성혈관능력유관。결론신타미신촉진료손상혈관내피적재내피화진정。
Objective To study the role and mechanism of indomethacin on the damaged carotid artery re-endothelialization .Meth-ods Carotid artery balloon injury models of SD rats were made , intraperitoneal injection of indomethacin , then the impact of Evans blue staining on endothelial repair and density gradient centrifugation of rat 's monocytes were observed;endothelial progenitor cells in vitro were cultured with EGM-2 medium.The late endothelial progenitor cells were treated with different concentrations of indomethacin after 24 h, the proliferation activity by MTT assay , the differences of EPCs adhesion were detected by cell adhesion methods , modified Boyden chamber de-tection of EPCs , Matrigel detection of EPCs into vascular capacity .Results Indomethacin significantly promoted the repair of injured vascu-lar endothelium, which might be related with enhanced proliferation , adhesion, migration and angiogenic ability of endothelial progenitor cells by indomethacin .Conclusions Indomethacin promots the process of re-endothelialization to the injured vascular endothelial .
