目的:对九里香血清药物化学抗软骨细胞凋亡进行初步研究。方法建立大鼠骨关节炎( OA)模型,含药血清培养软骨细胞,利用流式细胞仪检测其凋亡,建立中药九里香的指纹图谱、血清指纹图谱等方法,利用柱层析从原药材中分离鉴定主要成分。结果分离培养软骨细胞,九里香含药血清可显著抑制OA软骨细胞的凋亡情况。在大鼠血清中存在16个入血成分,其中6个为九里香的原型成分,10个可能为原型成分的代谢产物。分离鉴定两个主要成分九里香酮和phebalosin,分别与血清中色谱峰9和11的相对保留时间基本一致。结论九里香含药血清具有抗OA软骨细胞凋亡作用,入血成分或移行成分可能成为九里香在体内的直接作用物质。

목적:대구리향혈청약물화학항연골세포조망진행초보연구。방법건립대서골관절염( OA)모형,함약혈청배양연골세포,이용류식세포의검측기조망,건립중약구리향적지문도보、혈청지문도보등방법,이용주층석종원약재중분리감정주요성분。결과분리배양연골세포,구리향함약혈청가현저억제OA연골세포적조망정황。재대서혈청중존재16개입혈성분,기중6개위구리향적원형성분,10개가능위원형성분적대사산물。분리감정량개주요성분구리향동화phebalosin,분별여혈청중색보봉9화11적상대보류시간기본일치。결론구리향함약혈청구유항OA연골세포조망작용,입혈성분혹이행성분가능성위구리향재체내적직접작용물질。
Objective To study anti-chondrocyte apoptosis of the serum pharmacochemistry of Murraya exotica .Methods Rat OA models were eatablished , chondrocyte cells were separated and cultured , the apoptotic rate of chondrocyte was tested by flow cytometer .Tra-ditional Chinese Medicine fingerprint and serum fingerprint of M .Exotica were established ,main constituents of M .Exotica was isolated and identified.Results The serum pharmacochemistry significantly decreased the apoptotic rate of OA chondrocyte .16 constituents were found in the rat serum,of which, 6 compounds were original from M .exotica, 10 might be the metabolites of the original constituents .Murrayone and phebalosin were identified as the two major compounds of M .exotica, which exhibited the same relative retention time as those in the se-rum.Conclusions The serum pharmacochemistry of M .exotica exhibits the activity of anti-chondrocyte apoptosis .The original constituents or the metabolites might be the compounds that are responsible for the biological activity of M .exotica directly .Further isolation and identifi-cation of the active compounds could better explain the therapeutic basis matters of M .exotica .
